Biodegradation in water
Biodegradability of Dipotassium 4,4'-bis[6-anilino-4-[bis(2-hydroxyethyl)amino-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonate (CAS no. 71230 -67 -6) is predicted using OECD QSAR toolbox version 3.3 (2017) with logKow as the primary descriptor. Test substance undergoes 1.55% degradation by BOD in 28 days. Thus, based on percentage degradation, the test chemical Dipotassium 4,4'-bis[6-anilino-4-[bis(2-hydroxyethyl)amino-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonate was estimated to be not readily biodegradable in water.
Biodegradation in water & sedimemt
Estimation Programs Interface (EPI Suite, 2017) prediction model was run to predict the half-life in water and sediment for the test compound Dipotassium 4,4'-bis[6 -anilino-4 -[bis(2 -hydroxyethyl)amino-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate (CAS No. 71230 -67 -6). If released in to the environment, 0.684% of the chemical will partition into water according to the Mackay fugacity model level III and the half-life period of Dipotassium 4,4'-bis[6-anilino-4-[bis(2-hydroxyethyl)amino-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonate in water is estimated to be 180 days (4320 hrs). The half-life (180 days estimated by EPI suite) indicates that the chemical is persistent in water and the exposure risk to aquatic animals is moderate to high whereas the half-life period of Dipotassium 4,4'-bis[6 -anilino-4 -[bis(2 -hydroxyethyl)amino-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate in sediment is estimated to be 1620.833 days (38900 hrs). Based on this half-life value, it indicates thatDipotassium 4,4'-bis[6 -anilino-4 -[bis(2 -hydroxyethyl)amino-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate is persistent in sediment.
Biodegradation in soil
The half-life period of Dipotassium 4,4'-bis[6-anilino-4-[bis(2-hydroxyethyl)amino-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonate (CAS No. 71230 -67 -6) in soil was estimated using Level III Fugacity Model by EPI Suite version 4.1 estimation database (EPI suite, 2017). If released into the environment, 40.2% of the chemical will partition into soil according to the Mackay fugacity model level III. The half-life period of Dipotassium 4,4'-bis[6-anilino-4-[bis(2-hydroxyethyl)amino-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonate in soil is estimated to be 360 days (8640 hrs). Based on this half-life value of Dipotassium 4,4'-bis[6-anilino-4-[bis(2-hydroxyethyl)amino-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonate, it is concluded that the chemical is persistent in the soil environment and the exposure risk to soil dwelling animals is moderate to high.
Bioaccumulation: aquatic/sediment
The bioaccumulation study was conducted in fish for estimating the BCF bioaccumulation factor) value of test chemical Dipotassium 4,4'-bis[6 -anilino-4 -[bis(2 -hydroxyethyl) amino-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate (CAS no. 71230 -67 -6). The BCF value was calculatedusing a SRC-BCFWIN v2.15, program and a log Kow of 0.65. The BCF (bioaccumulation factor) value of Dipotassium 4,4'-bis[6 -anilino-4 -[bis(2 -hydroxyethyl)amino-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate was estimated to be 3.162 dimensionless, which does not exceed the bioconcentration threshold of 2000, indicating that the chemical Dipotassium 4,4'-bis[6 -anilino-4 -[bis(2 -hydroxyethyl)amino-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate is considered to be non-accumulative in aquatic organisms.
Adsorption/desorption
Adsorption study was conducted for estimating the adsorption coefficient (Koc) value of test chemical Dipotassium 4,4'-bis[6 -anilino-4 -[bis(2 -hydroxyethyl)amino-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate (CAS no. 71230 -67 -6). The adsorption coefficient (Koc) value of test substance Dipotassium 4,4'-bis[6 -anilino-4 -[bis(2 -hydroxyethyl)amino-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonatewas estimated to be 8100000 L/Kg (Log Koc = 6.908). This Koc value indicates that the substance Dipotassium 4,4'-bis[6 -anilino-4 -[bis(2 -hydroxyethyl)amino-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate has a very strong sorption to soil and sediment and therefore have negligible migration potential to ground water.

In a supporting weight of evidence study from authoritative database (HSDB, 2017) for the read across chemical disodium 2,2'-ethene-1,2 -diylbis{5 -[(4 -anilino-6 -morpholin-4 -yl-1,3,5 -triazin-2 -yl)amino]benzenesulfonate} (CAS no. 16090 -02 -1), biodegradation experiment was carried out using river die away test for 35 days for evaluating the percentage biodegradability of read across substance disodium 2,2'-ethene-1,2 -diylbis{5 -[(4 -anilino-6 -morpholin-4 -yl-1,3,5 -triazin-2 -yl)amino]benzenesulfonate}. Initial test substance conc. used was 1 mg/l (1000 ppb). The percentage degradation of substance disodium 2,2'-ethene-1,2 -diylbis{5 -[(4 -anilino-6 -morpholin-4 -yl-1,3,5 -triazin-2 -yl)amino]benzenesulfonate} was determined to be 0% after 35 days by using CO2 evolution parameter. Thus, based on percentage degradation, disodium 2,2'-ethene-1,2 -diylbis{5 -[(4 -anilino-6 -morpholin-4 -yl-1,3,5 -triazin-2 -yl)amino]benzenesulfonate} is considered to be not readily biodegradable in nature.
For the same read across chemical disodium 2,2'-ethene-1,2-diylbis{5-[(4-anilino-6-morpholin-4-yl-1,3,5-triazin-2-yl)amino]benzenesulfonate} (CAS no. 16090-02-1), biodegradation study was conducted for 5 days for evaluating the percentage biodegradability of read across substance disodium 2,2'-ethene-1,2 -diylbis{5 -[(4 -anilino-6 -morpholin-4 -yl-1,3,5 -triazin-2 -yl)amino]benzenesulfonate} (HSDB, 2017). Bacteria was used as a test inoculum for the study. The 5 day BOD value of chemical disodium 2,2'-ethene-1,2 -diylbis{5 -[(4 -anilino-6 -morpholin-4 -yl-1,3,5 -triazin-2 -yl)amino]benzenesulfonate} was determined to be 0 g O2/g test material. Thus, based on this BOD5 value, substance disodium 2,2'-ethene-1,2 -diylbis{5 -[(4 -anilino-6 -morpholin-4 -yl-1,3,5 -triazin-2 -yl)amino]benzenesulfonate} is considered to be not readily biodegradable in nature.
In a supporting weight of evidence study from secondary source (OECD SIDS, 2005 and HPVIS, 2017) for the read across chemical 2,2'-ethene-1,2 -diylbis[5 -({4 -anilino-6 -[bis(2 -hydroxyethyl)amino]-1,3,5 -triazin-2 -yl}amino)benzenesulfonic acid] (CAS no. 4404 -43 -7), biodegradation experiment was conducted for 28 days for evaluating the percentage biodegradability of read across substance 2,2'-ethene-1,2 -diylbis[5 -({4 -anilino-6 -[bis(2 -hydroxyethyl)amino]-1,3,5 -triazin-2 -yl}amino)benzenesulfonic acid]. The study was performed according to OECD Guideline 301 D (Ready Biodegradability: Closed Bottle Test) under aerobic conditions. Activated sludge (domestic, non-adapted) was used as a test inoculum for the study. Reference substance aniline undergoes 81 and 83% degradation after 15 and 28 days respectively. The percentage degradation of substance 2,2'-ethene-1,2 -diylbis[5 -({4 -anilino-6 -[bis(2 -hydroxyethyl)amino]-1,3,5 -triazin-2 -yl}amino)benzenesulfonic acid] was determined to be 7, 8 and <10% by using O2 consumption parameter in 5, 10 and 28 days, respectively. Thus, based on percentage degradation, 2,2'-ethene-1,2 -diylbis[5 -({4 -anilino-6 -[bis(2 -hydroxyethyl)amino]-1,3,5 -triazin-2 -yl}amino)benzenesulfonic acid] is considered to be not readily biodegradable in nature.
Another biodegradation study was conducted for 28 days for evaluating the percentage biodegradability of read across substance disodium 4,4'-bis[6 -anilino-[4 -[bis(2 -hydroxyethyl)amino]-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate (CAS no. 4193 -55 -9) (OECD SIDS, 2005 and HPVIS, 2017).The study was performed according to OECD Guideline 303 A "Simulation Test - Aerobic Sewage Treatment: Coupled Unit Test" under aerobic conditions. Activated sludge obtained from sewage treatment plant (STP) was used as a test inoculum for the study. Initial test substance concentration used for the study was 117.6 mg/l based on TOC (20 mg/l) (TOC content of test substance: 17%). TOC content in nutrient solution was 115 mg/l. No reference substance was used for the study. The percentage degradation of substance disodium 4,4'-bis[6 -anilino-[4 -[bis(2 -hydroxyethyl)amino]-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate was determined to be 11% degradation by using TOC removal parameter in 21 days. Thus, based on percentage degradation, disodium 4,4'-bis[6 -anilino-[4 -[bis(2 -hydroxyethyl)amino]-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate is considered to be not readily biodegradable in nature.
For the read across chemicaltetrasodium 2,2'-ethene-1,2-diylbis[5-({4-[bis(2-hydroxyethyl)amino]-6-[(4- sulfonatophenyl)amino]-1,3,5-triazin-2-yl}amino)benzenesulfonate] (CAS no. 16470-24-9) from secondary source (OECD SIDS, 2001 and HPVIS, 2017), biodegradation experiment was conducted for 28 days for evaluating the percentage biodegradability of read across substance tetrasodium 2,2'-ethene-1,2-diylbis[5-({4-[bis(2-hydroxyethyl)amino]-6-[(4- sulfonatophenyl)amino]-1,3,5-triazin-2-yl}amino)benzenesulfonate]. The study was performed according toEPA OTS 796.3180 (Ready Biodegradability: Modified AFNOR Test) and OECD Guideline 301 A (Ready Biodegradability: DOC Die Away Test), respectively, under aerobic conditions. Domestic sewage effluent was used as a test inoculum obtained from sewage treatment plant. Initial test substance concentration used for the study was 40 mg/l based on DOC (Dissolved Organic Carbon). Modifications of the guideline involve sterilisation instead of sterile filtration and bacterial concentration of 2.6000000 per ml, respectively. The percentage degradation of substance tetrasodium 2,2'-ethene-1,2 -diylbis[5 -({4 -[bis(2 -hydroxyethyl)amino]-6 -[(4 - sulfonatophenyl)amino]-1,3,5 -triazin-2 -yl}amino)benzenesulfonate] was determined to be 1.2% by using DOC removal parameter in 28 days. Thus, based on percentage degradation, tetrasodium 2,2'-ethene-1,2 -diylbis[5 -({4 -[bis(2 -hydroxyethyl)amino]-6 -[(4 - sulfonatophenyl)amino]-1,3,5 -triazin-2 -yl}amino)benzenesulfonate] is considered to be not readily biodegradable in nature.
On the basis of above results for target chemical Dipotassium 4,4'-bis[6-anilino-4-[bis(2-hydroxyethyl)amino-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonate (from OECD QSAR toolbox version 3.3, 2017) and for its read across substance (from authoritative database and secondary source), it can be concluded that the test substance Dipotassium 4,4'-bis[6 -anilino-4 -[bis(2 -hydroxyethyl)amino-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate can be expected to be not readily biodegradable in nature.

In an experimental weight of evidence study from secondary source (OECD SIDS, 2005) and authoritative database (INERIS, 2009) for the target chemical Dipotassium 4,4'-bis[6-anilino-4-[bis(2-hydroxyethyl)amino-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonate (CAS No. 71230-67-6), bioaccumulation experiment was conducted in fish for estimating the BCF bioaccumulation factor) value of test chemical Dipotassium 4,4'-bis[6 -anilino-4 -[bis(2 -hydroxyethyl) amino-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate. For the read across chemical disodium 2,2'-ethene-1,2-diylbis{5-[(4-anilino-6-morpholin-4-yl-1,3,5-triazin-2-yl)amino]benzenesulfonate} (CAS no. 16090-02-1) from authoritative databases (J-CHECK, HSDB, 2017 and EnviChem, 2014),bioaccumulation study was conducted on test organism Cyprinus carpio for 6 weeks for evaluating the bioconcentration factor (BCF value) of read across substance disodium 2,2'-ethene-1,2-diylbis{5-[(4-anilino-6-morpholin-4-yl-1,3,5-triazin-2-yl)amino]benzenesulfonate}. The study was performed according to other guideline "Bioaccumulation test of a chemical substance in fish or shellfish" provided in "the Notice on the Test Method Concerning New Chemical Substances”. Cyprinus carpio was used as a test organism for the study. Details on analytical methods involve the recovery ratio: Test water: 1st concentration area : 94.6%, 2nd concentration area : 91.3%, Fish: 85.9%, - Limit of quantitation: Fish : 0.093 ppm. Test chemical nominal conc. used for the study were 0.2 mg/land 0.02 mg/l, respectively. Dimethyl sulfoxide was used as a vehicle in the study. Range finding study involve the TLm (48 hr) ≥ 50 ppm (w/v) on Rice fish (Oryzias latipes).The bioconcentration factor (BCF value) of substance disodium 2,2'-ethene-1,2-diylbis{5-[(4-anilino-6-morpholin-4-yl-1,3,5-triazin-2-yl)amino]benzenesulfonate} on Cyprinus carpio was determined to be in the range of 1.4 – 4.7 L/Kg at a conc. of 0.2 mg/l and ≤ 6.4 – 28 L/Kg at a conc. of 0.02 mg/l, respectively.
In a supporting weight of evidence study from authoritative database (HSDB, 2017) for the read across chemical Benzenesulfonic acid,2,2'-(1,2-ethenediyl)bis[5-[[4-[(2-hydroxyethyl)methylamino]-6-(phenylamino)-1,3,5-triazin-2-yl]amino]-,sodium salt (1:2) (CAS no.13863-31-5), bioaccumulation experiment was conducted for determining the BCF (bioaccumulation factor) value of read across chemical Benzenesulfonic acid,2,2'-(1,2 -ethenediyl)bis[5 -[[4 -[(2 -hydroxyethyl)methylamino]-6 -(phenylamino)-1,3,5 -triazin-2 -yl]amino]-,sodium salt (1:2). The BCF (bioaccumulation factor) value of Benzenesulfonic acid,2,2'-(1,2 -ethenediyl)bis[5 -[[4 -[(2 -hydroxyethyl)methylamino]-6 -(phenylamino)-1,3,5 -triazin-2 -yl]amino]-,sodium salt (1:2) was determined to be < 2.1 dimensionless.
On the basis of above results for target chemical Dipotassium 4,4'-bis[6-anilino-4-[bis(2-hydroxyethyl)amino-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonate (from secondary source) and for its read across substance (from authoritative databases J-CHECK, HSDB, 2017 and EnviChem, 2014), it can be concluded that the BCF value of test substance Dipotassium 4,4'-bis[6-anilino-4-[bis(2-hydroxyethyl)amino-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonate was estimated to be 3.162 which does not exceed the bioconcentration threshold of 2000, indicating that the chemicalDipotassium 4,4'-bis[6-anilino-4-[bis(2-hydroxyethyl)amino-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonateis not expected to bioaccumulate in the food chain.

In an experimental weight of evidence study from authoritative database (INERIS, 2009) for the target chemical Dipotassium 4,4'-bis[6-anilino-4-[bis(2-hydroxyethyl)amino-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonate (CAS No. 71230-67-6), adsorption experiment was conducted for estimating the adsorption coefficient (Koc) value of test chemical Dipotassium 4,4'-bis[6 -anilino-4 -[bis(2 -hydroxyethyl)amino-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate. The adsorption coefficient (Koc) value of test substance Dipotassium 4,4'-bis[6 -anilino-4 -[bis(2 -hydroxyethyl)amino-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate was estimated to be 8100000 L/Kg (Log Koc = 6.908).
Another adsorption study was conducted for estimating the adsorption coefficient (Koc) value of test chemical Dipotassium 4,4'-bis[6 -anilino-4 -[bis(2 -hydroxyethyl)amino-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate (CAS no. 71230 -67 -6) (OECD SIDS, 2005). The adsorption coefficient (Koc) value was calculated using a PCKOCWIN v.1.66 program. The adsorption coefficient (Koc) value of test substance Dipotassium 4,4'-bis[6 -anilino-4 -[bis(2 -hydroxyethyl)amino-1,3,5 -triazin-2 -yl]amino]stilbene-2,2'-disulphonate was estimated to be 8133000000 (Log Koc = 9.91).
For the read across chemical Benzenesulfonic acid,2,2'-(1,2-ethenediyl)bis[5-[[4-[(2-hydroxyethyl)methylamino]-6-(phenylamino)-1,3,5-triazin-2-yl]amino]-,sodium salt (1:2) (CAS no. 13863-31-5) from authoritative database (HSDB, 2017), adsorption study was conducted for estimating the adsorption coefficient (Koc) value of read across chemical Benzenesulfonic acid,2,2'-(1,2 -ethenediyl)bis[5 -[[4 -[(2 -hydroxyethyl)methylamino]-6 -(phenylamino)-1,3,5 -triazin-2 -yl]amino]-,sodium salt (1:2). The adsorption coefficient (Koc) value was calculated using a structure estimation method based on molecular connectivity indices. The adsorption coefficient (Koc) value of substance Benzenesulfonic acid,2,2'-(1,2 -ethenediyl)bis[5 -[[4 -[(2 -hydroxyethyl)methylamino]-6 -(phenylamino)-1,3,5 -triazin-2 -yl]amino]-,sodium salt (1:2) was estimated to be 600000000 (Log Koc = 8.778).
On the basis of above overall results for target chemical Dipotassium 4,4'-bis[6-anilino-4-[bis(2-hydroxyethyl)amino-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonate(from authoritative database INERIS, 2009 and secondary source OECD SIDS, 2005) and for its read across substance (from authoritative database HSDB, 2017), it can be concluded that the Koc value of test substanceDipotassium 4,4'-bis[6-anilino-4-[bis(2-hydroxyethyl)amino-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonateranges from 8100000–8133000000 indicating that the test chemical Dipotassium 4,4'-bis[6-anilino-4-[bis(2-hydroxyethyl)amino-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonate has a very strong sorption to soil and sediment and therefore have negligible migration potential to ground water.
