Skin irritation:

Skin irritation test of test chemical 6-aminonaphthalene-1,3-disulfonic acid (CAS No: 118-33-2) was conducted on rabbit to examine the degree of skin irritancy caused by the chemical. The rabbits were exposed to 2 mg of chemical dermally for 24 hours and later observed for skin reactions. Since the chemical induce sever skin reactions,6-amino-1,3- Naphthalene disulfonic acid(CAS No: 118-33-2) was considered to be irritating to rabbits’ skin.

 

Eye irritation:

The eye irritation potential of 6-amino-1,3- Naphthalene disulfonic acid (CAS No: 118-33-2) was assessed in rabbits. 750 µg of 6-amino-1,3- Naphthalene disulfonic acid was instilled into rabbit eyes and observed for signs of irritation till 24 hours. 6-amino-1,3- Naphthalene disulfonic acid was severely irritating to rabbit eyes after 24 hours of exposure. Hence, 6-amino-1,3- Naphthalene disulfonic acid (CAS No: 118-33-2) can be considered to be an eye irritant.

The studies are based on in vivo experiments in rabbits for target chemical6-aminonaphthalene-1,3-disulfonic acid (CAS No: 118-33-2) and its structurally similar read across substances2-aminonaphthalene-1,5-disulfonic acid (CAS No: 117-62-4)and 7-aminonaphthalene-1,3,6-trisulphonic acid (CAS No: 118-03-6) .The predicted data using the OECD QSAR toolbox andDanish QSAR databasehas also been compared with the experimental data and summarized as below;

BAYER (1994) and RTECS (1997) to examine the degree of skin irritancy caused by the chemical on rabbit. In a prediction done by SSS (2017) using the OECD QSAR toolbox with log kow as the primary descriptor, the skin irritation potential was estimated for test chemical6-aminonaphthalene-1,3-disulfonic acid (CAS No: 118-33-2).The chemical 6-aminonaphthalene-1,3-disulfonic acid (CAS No: 118-33-2)is estimated to be irritating to skin of rabbits.

 

 

According toDanish QSAR database, theskin irritation effects were estimated by using four different models i.e, Battery, Leadscope, SciQSAR and CASE Ultra for Based on estimation, severe skin irritation effects were known when6-aminonaphthalene-1,3-disulfonic acid (CAS No: 118-33-2)was exposed to rabbit skin.

 

 

The above results were further supported by the experimental studies carried out by Richard J. Lewis, Sr. (2012)and RTECS (2015) for two structurally similar read across substances 7-aminonaphthalene-1,3,6-trisulphonic acid (CAS No: 118-03-6) and 2-aminonaphthalene-1,5-disulfonic acid (CAS No: 117-62-4) on rabbits’ skin. The rabbits were exposed to both the chemicals (individually) for 24 hrs at 500 mg and observed for signs of irritation. The treated rabbits produced mild irritating effects in both the studies. Thus the chemicals 7-aminonaphthalene-1,3,6-trisulphonic acid (CAS No: 118-03-6) and 2-aminonaphthalene-1,5-disulfonic acid (CAS No: 117-62-4) were considered as irritating to the skin of rabbits.

 

 

Thus on the basis of available data for the 6-aminonaphthalene-1,3-disulfonic acid (CAS No: 118-33-2) and its structurally similar read across substances2-aminonaphthalene-1,5-disulfonic acid (CAS No: 117-62-4)and 7-aminonaphthalene-1,3,6-trisulphonic acid (CAS No: 118-03-6) ,it can be concluded thatchemical 6-aminonaphthalene-1,3-disulfonic acid (CAS No: 118-33-2) is able to cause skin irritation and thus considered as irritating.Comparing the above annotations with the criteria of CLP regulation, it can be classified under the category “Category 2 (irritant)”.

In case of 2-aminonaphthalene-1,5-disulfonic acid (CAS No: 117-62-4),20 mg of 2-aminonaphthalene-1,5-disulfonic acid was instilled into the eyes of rabbits. Then rabbits were observed for signs of ocular lesions after 24 hours.

Both the chemicals caused moderate irritation to the rabbit eyes after 24 hours. Hence the chemicals 7-Amino-1,3,6-naphthalenetrisulfonic acid  (CAS No: 118-03-6) and 2-aminonaphthalene-1,5-disulfonic acid (CAS No: 117-62-4) can be considered as moderately irritating to the rabbits’ eye.
