There are no reliable aquatic toxicity data available for 2-(3,4-epoxycyclohexyl)ethyltrimethoxysilane (CAS 3388-04-3), therefore good quality data for the structurally-related substance, [3-(2,3-epoxypropoxy)propyl]diethoxy(methyl)silane (CAS 2897-60-1), have been read across.

The test substance is susceptible to two separate hydrolysis reactions of its different functional groups, as described in Section 5.1.2 of IUCLID. The chemical safety assessment focusses on the silanol hydrolysis product 2-(3,4-epoxycyclohexyl)ethylsilanetriol.

The available aquatic ecotoxicity studies conducted with the structurally-related substance are consistent with this and also indicate the formation of precipitated condensation products at longer time scales. Therefore it is likely that the test organisms were exposed to a mixture of the parent test substance and its hydrolysis products.

Reliable short-term toxicity tests results are available for freshwater fish (Oncorhynchus mykiss), invertebrates (Daphnia magna) and algae (Scenedesmus subspicatus) for [3-(2,3-epoxypropoxy)propyl]diethoxy(methyl)silane (CAS 2897-60-1). The relevant short-term values are:

Fish: LC50 (96 h): 139 mg/l; NOEC: 77 mg/l (expressed in terms of [3-(2,3-epoxypropoxy)propyl]methylsilanediol)

Daphnia: EC50 (48 h): 15.5 mg/l; NOEC: 1.4 mg/l (expressed in terms of [3-(2,3-epoxypropoxy)propyl]methylsilanediol)

Algae: EC50 (72 h): >25 mg/l; NOEC: 2.5 mg/l  (expressed in terms of [3-(2,3-epoxypropoxy)propyl]methylsilanediol).

There are no reliable microorganism toxicity data available for 2-(3,4-epoxycyclohexyl)ethyltrimethoxysilane (CAS 3388-04-3), therefore good quality data for the structurally-related substance, [2-(3,4-epoxycyclohexyl)ethyl]triethoxysilane (CAS 010217-34-2), have been read across. Both substances hydrolyse in the time scale of the test and have a structurally identical silanol hydrolysis products, 2-(3,4-epoxycyclohexyl)ethylsilanetriol.

An activated sludge respiration inhibition 30-minute NOEC value of 100 mg/l (nominal) was determined in a reliable study.

2-(3,4-epoxycyclohexyl)ethylsilanetriol is susceptible to further hydrolysis reactions and the ultimate hydrolysis product 2-(3,4-dihydroxycyclohexyl)ethylsilanetriol is considered unlikely to exhibit significant ecotoxic effects based on QSAR estimated E(L)C50 values >>100 mg/l (ECOSAR). The rate of reaction under environmental conditions is uncertain but the half-life is not expected to be more than a few days.
