Short-term toxicity to fish:
Short term toxicity to fish was assessed for the target compound (5or8)-aminonaphthalene-2-sulphonic acid (CAS no 51548-48-2) and its related read- across by reviewing a number of studies. The summary of the results are presented below:
Short term aquatic toxic effects of the test compound (5or8)-aminonaphthalene-2-sulphonic acid (CAS no 51548-48-2) was predicted (SSS QSAR, 2016). The study was based on the effects of the test compound on the fish species Brachydanio rerio (new name: Danio rerio) in 96 hrs duration in a fresh water static system. The predicted data suggests the lethal concentration (LC50) for (5or8)-aminonaphthalene-2-sulphonic acid on the basis of mortality to be 176.1891 mg/L. This value concluded that the test compound (5or8)-aminonaphthalene-2-sulphonic acid does not exhibit short term toxicity to fish (Brachydanio rerio (new name: Danio rerio)) and is non hazardous to the aquatic environment.
Other predicted model ECOSAR Version 1.10 indicate that, the 96 hours LC50 was estimated to be 74690.617mg/l on fish for substance (5or8)-aminonaphthalene-2-sulphonic acid with mortality effects.
Whereas experimental study (Chemophere. Vol. 28, No. 12, pp. 2203.2236, 1994) for read across substance 6-aminonaphthalene-1,3-disulphonic acid (118-33-2)indicate that the lethal concentration (LC0) of test substance for fish Leuciscus idus, Bruchydunio rerio,Salmo gaidneri, Pimephales promelas, Lepomis macrochirus, Poecilia reticulate was found to be 1000 mg/l for 96 hrs.
And other database (Data bank of Environmental Properties of Chemicals (EnviChem), 2014) for read across Sodium naphthionate (Cas no.130-13-2)indicate that the median lethal concentration (LC50) of test substance substance Sodium naphthionate for fish Oryzias latipes was found to be >4000 mg/l for 48 hrs. on the basis of mortality effects.
Based on above available studies it is concluded that the substance 5or8)-aminonaphthalene-2-sulphonic acid (CAS no 51548-48-2) not qualify for the aquatic classification as per the CLP criteria.
Short term toxicity to aquatic invertebrates:
Various predicted studies for the (5Or8)-aminonaphthalene-2-sulphonic acid were reviewed to summarize the following information.
48 hrs aquatic toxicity studies (SSS QSAR prediction model, 2016) were conducted to assess toxic effects of the (5Or8)-aminonaphthalene-2-sulphonic acid (CAS no 51548-48-2) and the results were predicted. The study was based on the effects of the test compound on the Daphnia magna in a static fresh water system. The predicted data suggests the effective concentration (EC50) for the (5Or8)-aminonaphthalene-2-sulphonic acid was estimated to be 111.585372925 mg/L on the basis of mobility.
48 hrs aquatic toxicity study (Danish QSAR, 2016) was conducted to assess toxic effects of the (5Or8)-aminonaphthalene-2-sulphonic acid (CAS no 51548-48-2) and the results were predicted. The study was based on the effects of the test compound on Daphnia magna in a static fresh water system. The predicted data suggests the effective concentration (EC50) for the (5Or8)-aminonaphthalene-2-sulphonic acid was estimated to be 307.1647 mg/l.
48 hrs aquatic toxicity studies (EPI suite, ECOSAR version 1.1, 2016) were conducted to assess toxic effects of the (5Or8)-aminonaphthalene-2-sulphonic acid (CAS no 51548-48-2) and the results were predicted. The study was based on the effects of the test compound on the Daphnia magna in a static fresh water system. The predicted data suggests the lethal concentration (LC50) for the (5Or8)-aminonaphthalene-2-sulphonic acid was estimated to be 32429.133 mg/Lon the basis of mobility.
On the basis of results for toxicity to aquatic invertebrates, it can be considered that the substance is not likely to be toxic to aquatic invertebrates at environmentally relevant concentrations and also can be classified as non- hazardous as per the criteria of CLP regulation.
Toxicity to aquatic algae and cyanobacteria
Various predicted data for the test chemical(5Or8)-aminonaphthalene-2-sulphonic acid(CAS No. 51548-48-2)were reviewed to summarize the following information:
72 hrs aquatic toxicity study was predicted(SSS QSAR Prediction model, 2016)to assess toxic effects of the test compound (5Or8)-aminonaphthalene-2-sulphonic acid (CAS no. 51548 -48 -2) and the result were predicted. The study was based on the effects of the test compound on Desmodesmus subcapitata in a static fresh water system. The predicted data suggests the effective concentration (EC50) for the test compound (5Or8)-aminonaphthalene-2-sulphonic acid (CAS no.51548 -48 -2) was estimated to be 110.384 mg/l on the basis of growth rate.
Based on the QSAR prediction done using the EPI Suite ECOSAR version 1.10 (EPI Suite, 2016), the 96 hours EC50 was estimated to be 7965.639 mg/l on green algae for substance (5Or8)-aminonaphthalene-2-sulphonic acid with growth inhibition effects.
Estimated 72 hrs EC50 value (Danish QSAR database, 2016) of test substance (5Or8)-aminonaphthalene-2-sulphonic acid on Pseudokirchneriella subcapitata was determined by three different models i.e, Battery, Leadscope and SciQSAR used within Danish QSAR database. Based on inhibition in growth rate of test organismPseudokirchneriellasubcapitata, the estimated 72 hrs EC50 value was found to be 2728.812 mg/l.
Based on the overall reported results for test substance, it can be concluded that the substance(5Or8)-aminonaphthalene-2-sulphonic acid can be considered as non-toxic to aquatic organisms and thus cannot be classified as hazardous as per the CLP criteria.
