PREPARATION AND APPLICATION OF TEST SOLUTION
Preparation of the saturated solution
The saturated solution (1.00 mg/L test item was weighed out) was prepared with dilution water two days prior to the start of exposure (0 h).

Treatment
The stock solution was stirred for 48 hours (1100 rpm) with a magnetic stirrer. Undissolved particles were removed by membrane filtration (membrane filter 0.45 µm, RC, Macherey-Nagel).

Control
7 fish in dilution water (without test item) were tested under the same test conditions as the test replicates.

TEST ORGANISM
- Common name: Zebrafish
- Source: All fish used in the test were gained at DR.NOACK-LABORATORIEN from a single brood stock. (supplier: Niedersächsischer Landesbetrieb für Wasserwirtschaft, Küsten- und NAturschutz, An der Scharlake 39, D-31135 Hildesheim)
- Length at the test end (length definition, mean, range and SD): Average body length: 2.36 cm
- Weight at the test end (mean and range, SD): Average body weight: 0.156 g
- Method of breeding:Holding was performed at the test facility at 23 +/- 2 °C and diffuse light (0.1 - 10 µmol photons • m-2 • s-1, natural photoperiod). The water was changed at least once per week. The dissolved oxygen concentration was more than 80 % of the air saturation value. No disease treatments were administered throughout holding and testing. Food was provided 3 times per week. The amount of food was 4 % of the fish body weight per feeding day. The test fish were not fed 24 h before the test started.

ACCLIMATION
- Acclimation period: Zebrafish with at least 12 days of acclimatisation and mortality < 5 % within these days before the study starts were used in the test.

TEST MEDIUM / WATER PARAMETERS
- Source/preparation of dilution water: Tap water of local origin was used for holding and testing. The water was filtered on activated charcoal and aerated for at least 24 h to remove possible chlorine.

EFFECT PARAMETERS MEASURED (with observation intervals if applicable) : Fish were considered dead if there was no visible movement (e.g. gill covers movement) and if touching of the caudal peduncle produced no reaction. Records were kept of visible abnormalities (e.g. loss of equilibrium, swimming behaviour, respiratory function, pigmentation, etc.). Observations were made after 24, 48, 72 and 96 h.

Conclusions:
In this study, the saturated solution of the test item was found not to cause lethal effects to zebrafish after 96 hours at a nominal concentration of 1.00 mg/L. The LC50 after 96 hours was > 1.00 mg/L, the LC0 was 1.00 mg/L.
Executive summary:

The acute toxicity of the test item (batch no. USEA016274) to fish (zebrafish) was determined according to the principles of the OECD-Guideline for Testing of Chemicals No. 203 (1992) and EU Council Regulation (EC) No. 440/2008/ Method C.1 (2008) .

 

A static test was conducted with a saturated solution prepared from a nominal concentration of 1.00 mg/L. The duration of the test was 96 hours. 7 test organisms each were exposed to the limit concentration and the control, respectively. The water quality parameters temperature, pH-value and oxygen saturation measured at 0, 24, 48, 72 and 96 hours were determined to be within the acceptable limits.

 

The concentrations of the test item were not determined analytically because no suitable method for the determination of the test item could be established. All effect values are presented based on the nominal test item concentration.

Description of key information

The acute toxicity of the test item to fish (zebrafish) was determined according to OECD-Guideline for Testing of Chemicals No. 203 (1992). A static limit test was conducted with a saturated solution prepared from a nominal concentration of 1.00 mg/L of the test item.


The saturated solution of the test item didn´t show any signs of toxicity nor lethality at all observation time points up to 96 hours. Therefore, the NOEC and LC0were laid down as 1.00 mg/L (based on the nominal concentration) and LC100was laid down as > 1.00 mg/L (based on the nominal concentration).
