Description of key information

Based on the nominal concentrations of the test item Acid Red EAY 9656, the 48 hour-EC50 for Daphnia magna was 125 mg/L  (95 % confidence limits: 108 – 151 mg/L). The NOEC after 48 hours was 50.0 mg/L. The LOEC after 48 hours was 100 mg/L.

Additional information

In the acute immobilisation test withDaphnia magna (STRAUS), the effects of the test item Acid Red EAY 9656 were determined according to OECD 202 (2004), equivalent to the Council Regulation (EC) No.440/2008 Method C.2 (2008).

The study was conducted under static conditions over a period of 48 hours with 5 nominal concentrations of the test item Acid Red EAY 9656 in the range of 12.5 - 200 mg/L, prepared with dilution water (ISO test water, acc. to OECD202, Annex 3) in a geometric series with a separation factor of 2.

The test item Acid Red EAY 9656 is a dark red powder. A stock solution of 200 mg/Lof the test item was freshly prepared with dilution water before the start of the exposure (at 0 hours), treated with ultrasound for 15 minutes at room temperature, followed by stirring with approximately 1100 rpm for 15 minutes at room temperature and used for the preparation of all further concentration levels and as highest test concentration.The tested concentration levels were visually clear and showed a concentration related red colour throughout the exposure period.

Twenty daphnids were exposed to each concentration level and the control.

The concentrations of the test item Acid Red EAY 9656wereanalytically verified via HPLC-DAD at the start (0 hours) and at the end of the exposure (48 hours) in all concentration levels and the control.

The measured concentrations of the test item at the start of the exposure (0 hours) were in the range of 103 to 113 % of the nominal concentrations and 95 to 103 % at the end of the exposure (48 hours).

The measured test item concentrations were all within ± 20 % of the nominal concentrations. This indicates that the test item concentrations were successfully maintained for the duration of the test. Therefore, the effect levels were based on the nominal concentrations of the test item Acid Red EAY 9656.

The water quality parameters (i.e. pH-value and dissolved oxygen concentration), measured at the start (0 hours) and at the end of the exposure (48 hours), were within the acceptable limits. The validity criteria of the test guidelines were fulfilled.
