The water solubility of the substance was determined according to a method equivalent or similar to OECD TG105 and EU Method A.6 as a part of a guideline OECD TG 201 algal growth inhibition test and in accordance with OECD 23 (2000) guidance. For each loading level, an appropriate amount of the test item was weighed out and transferred into a brown glass flask with an appropriate amount of demineralized water. Prior to environmental toxicity testing initiation: concentrations of test substance were analytically verified via analysis of total organic carbon (TOC, according to DIN EN 1484) at the start of the exposure (fresh solutions without components of the dilution water, 0 hours). The method generates the loading rate dependent water solubility based on total organic carbon (TOC) measurement. Analysis of the TOC is the most suitable method for the analytical monitoring of poorly soluble substances. The headspace in the brown glass flasks was minimized. For this purpose, the brown glass flask were nearly filled up. These dispersions were shaken for 24 hours with 20 rpm at room temperature. After a separation phase of at least 24 hours, the aqueous phase or WAFs were removed from the approximate bottom of the glass flask. The WAFs were checked via laser beam (Tyndall effect) for undissolved test item (formation of an emulsion). After the sampling for TOC measurement the components of the dilution water (test medium) were added to the WAFs. For each loading level, an appropriate amount of the test item was weighed out and transferred into a brown glass flask with an appropriate amount of demineralized water. Five water accommodated fractions (WAFs) were prepared with nominal loadings of the test item in a geometric series with a factor of 2: 1.25- 2.50- 5.00- 10.0- 20.0 mg/L. All water accommodated fractions (in demineralized water, without components of the dilution water (test medium)) were analytically verified. The test substance water solubility as a function of loading rate of a water accommodated fraction (WAF) was determined by Total Organic Carbon (TOC) measurement to be: nominal WAF: TOC measurement respectively: 20.0 mg/L : 3.77 mg C/L; 10.0 mg/L: 2.79 mg C/L and at 5.0 mg/L nominal WAF or below - the TOC was < 2.0 mg C/L at solution pH 7 and temperature 21.8 ± 0.5°C.
