Biodegradation in water: screening tests

Description of key information

Biodegradation in water ability was predicted for the test compound4-Amino-5-hydroxynaphthalene-1,7-disulphonic acid (CAS No. 130-23-4) using the SSS QSAR prediction model. The ready biodegradability of the test substance 4-Amino-5-hydroxynaphthalene-1,7-disulphonic acid was estimated as 26.7999% by using O2 consumption parameter. This result indicates that 4-Amino-5-hydroxynaphthalene-1,7-disulphonic acidwas estimated to be not readily biodegradable in water.

The predicted data for the test compound 4-Amino-5-hydroxynaphthalene-1,7-disulphonic acid (CAS No. 130-23-4) and its read across substance studies were reviewed for the biodegradation end point which are summarized as below:

Estimation Programs Interface Suite (EPI suite) was run to predict the biodegradation potential of the test compound4-Amino-5-hydroxynaphthalene-1,7-disulphonic acid (CAS No. 130-23-4). The screening test inherent to the biodegradability of the substance was calculated using the software BIOWIN v4.10. The results indicate4-Amino-5-hydroxynaphthalene-1,7-disulphonic acidwas estimated to be not readily biodegradable in water.

 

Biodegradation of substance 2-Amino-8-naphthol-6-sulfonic acid in water was determined (J-check study report, 2010). The test substance was found to be 5.3 % biodegraded in water after 2 wk on the basis of O2 consumption, BOD. 0.4% by TOC removal and <0 % by (UVVIS) parameters. Hence, the 2-Amino-8-naphthol-6-sulfonic acid is considered to be not biodegradable in water.

A closed bottle test according to OECD 301D guidelines (GREIM H., et. al; 1994) was performed to determine the ready biodegradability of 7-Amino-4 hydroxy- 2 – naphthalenesulphonic acid. No degradation of test chemical 7-Amino-4 hydroxy- 2 – naphthalenesulphonic acid was observed. Therefore, the chemical was considered to be not readily biodegradable.

Biodegradation screening test (J-CHECK, 2016) was conducted for 14 days (2 weeks) for evaluating the percentage biodegradability of the test substance.

Concentration of inoculum i.e, sludge is 30 mg/l and initial test substance conc. used in the study was 100 mg/l. The percentage degradation of test substance was found to be 6.3% by BOD, 8.4% by TOC and 0% by UV-VIS Thus, the substance 7-Amino-4 hydroxy- 2 - naphthalenesulphonic acid was considered to be not readily biodegradable in water.

Biodegradation screening test [Data bank of Environmental Properties of Chemicals (EnviChem), 2014] was conducted for 14 days for evaluating the percentage biodegradability of the test substance.

On the basis of above results of various studies and predicted data for target and read across substance, it can be concluded that the substance can be considered as non-biodegradable.
