A GLP guideline study according to OECD 221 using Lemna gibba as a test organism was conducted (BASF SE 99E0861/11X565, 2012). This test was conducted instead of a study using algae because the test substance is a dyestuff which would influence the photosynthesis capacity of algae cells in the test solution. Test results using other dye stuffs indicate that the toxicity level of dye stuffs is mainly determined by the shading effect of the test substance in the test system, thereby inhibiting the photosynthesis capacity of the algae.

By using the Lemna test, direct toxicity of the substance is established without interference of the light filtering effect of the test substance in the test system.

In this study, the ErC50 (7d) values on the frond number and the dry weight of the plants were determined to exceed the analytically verified test concentration of 320 mg/L. The lowest ErC10 (7d) was determined to be 2.02 mg/L for the dry weight of the plants.
