The solutions have to be prepared just before the test. Together with the blank, a series of solutions with concentration at a geometric series, to obtain 100% mortality after 48h and intermediate concentration to obtain a realistic EC50 at 48h.
The protocol ISO 147 has been followed, saturated solutions have been prepared at different concentrations. After 24 stirring, the fraction is stabilised for 4 hours and only the fraction with stables solution and emultion is used for the test. Since the toxicity could be generated both by the soluble fraction and the emulsion, a filtration is not appropriate.
