Description of key information

The acute toxicity of 4-ADPA to aquatic invertebrates (Dapnia magna) was conducted in a semistatic immobilisation test according EPA OTS 797.1300 (Aquatic Invertebrate Acute Toxicity Test, Freshwater Daphnids). After 48 hours of exposure an EC50 of 0.29 mg/L (nominal) is obtained (VKI, 2002).

OECD SIDS gave VKI-2002 a reliability 1 in 2005 in consideration of analytical monitoring carried out. However, this study only analyzed the concentration of 4 -ADPA in the stock solution and further estimated the actual concentration in the test vessels. Therefore, a reliability of 2 is appointed to this study in the up-to-date assessment. An EC50 -48h was estimated from linear regression and reported as 0.37 mg/l in the original publication (VKI, 2002). However, after updated calculation, the correct value should be 0.29mg/l. The estimation of effective concentration in OECD SIDS for this study with hydrolysis half-life time is also inappropriate, because a refreshment of test solution after 24h was carried out and no significant hydrolysis can be expected to happen in 24h (BIS, 2004).
