2% in the diet: increase in liver weights and hepatic peroxisomal enzyme activities (carnitine acetyltransferase and catalase), non-adverse

A study investigating peroxisome proliferation in the liver is available, in which 4 male Fischer 344 rats received Bis(2-ethylhexyl) sebacate at a concentration of 2% in the diet for 21 consecutive days (Moody, 1978). An additional group of 13 animals was fed the plain diet and served as control. At necropsy, a statistically significant increase in liver weights and hepatic peroxisomal enzyme activities (carnitine acetyltransferase and catalase) was observed in treated animals compared to controls. Furthermore, a statistically significant, but not specified decrease in serum triglycerides compared to controls was reported, whereas serum cholesterol remained unchanged. In conclusion, the substance orally administered at 2% in the diet caused hepatic peroxisomal proliferation, which was considered to be an adaptive response and not necessarily an adverse effect. However, the biological implication of peroxisome proliferating activity of environmental contaminants remains to be determined.
