Description of key information

Guideline study with no deviations and compliant with GLP. The octanol-water partition coefficient (Pow) for Modified Small Vinyl Ester was determined using the HPLC method at neutral pH and at pH = 1.5. Due to its complex composition, Pow was determined only for the two major constituents (bisGMA, ca. 57.5%; and monomaleic bisGMA, ca. 14.5%). At neutral pH logPow for monomaleic bisGMA and bisGMA was determined to <0.3 and 4.1, respectively; at pH = 1.5 logPow was determined to 4.3 for both constituents.

The octanol-water partition coefficient of Modified Small Vinyl Ester was determined in a guideline study using the HPLC method at neutral pH and at pH = 1.5. Due to complex composition of Modified Small Vinyl Ester, Pow was determined only for the two major constituents (bisGMA, ca. 57.5%; and monomaleic bisGMA, ca. 14.5%). In addition to the guideline study, the octanol-water partion coefficients of the two main constituents of Modified Small Vinyl Ester (bisGMA and monomaleic bisGMA) were calculated using the Rekker method.

The HPLC chromatograms showed two major peaks and a number of small peaks. The major peaks were allocated to the main constituents of Modified Small Vinyl Ester (bisGMA and monomaleic bisGMA). The small peaks were allocated to other constituents and to the oligomeric substances composing the third constituent. At neutral pH logPow for monomaleic bisGMA and bisGMA was determined to <0.3 and 4.1, respectively; at pH = 1.5 logPow was determined to 4.3 for both constituents. The calculated logPow's were 6.8 and 7.9 for monomaleic bisGMA and bisGMA, respectively.

Based on these experimental data, logPow for Modified Small Vinyl Ester is set to 4.3 at neutral pH.
