Description of key information

The substance FAT 40841/A TE shows a complex behaviour with regard to hydrolysis, depending on the temperature and the pH conditions of the environment. Overall, it can be concluded that the substance tends to be relatively stable in water under environmental conditions. The hydrolysis half-life was about 4 days under neutral pH conditions at 25 °C and increased considerably at pH 4 and pH 9.

The hydrolysis behaviour of the substance FAT 40841/A TE was tested in accordance with the EU Method C.7 and GLP at 50 °C at pH 4.0, 7.0 and 9.0. The substance showed under the test conditions three test item peaks in the HPLC chromatogram.

At pH 4.0, FAT 40841/A TE showed three main peaks in the chromatogram. The first and second test item peaks increased. Therefore, no hydrolysis behavior could be observed at 25 °C. As the third test item peak was not stable, further testing was performed at elevated temperatures in order to calculate the rate constant (k25) and the half-life time of the hydrolysis at pH 4.0 at 25 °C.

At pH 7.0, FAT 40841/A TE showed three main peaks in the chromatogram. The first test item peaks increased. Therefore, no hydrolysis behavior could be observed at 25 °C. As the second test item peak was not stable, further testing was performed at elevated temperatures in order to calculate the rate constant (k25) and the half-life time of the hydrolysis at pH 7.0 at 25 °C. The third test item peak was unstable at pH 7.0 and 50 °C. Therefore, no further testing was performed at this pH-value for this peak.

At pH 9.0, FAT 40841/A TE showed three main peaks in the chromatogram. The second test item peaks was unstable at pH 9.0 and 50 °C. Therefore, no further testing was performed at this pH value for this peak. The first and third test item peaks were not stable at pH 9.0 and 50 °C. Due to the hydrolysis behavior of the compounds of FAT 40841/A TE, the main test was performed only for the relevant peak at each pH. Therefore, further testing was performed for the third test item peak at elevated temperatures in order to calculate the rate constant (k25) and the half-life time of the hydrolysis at pH 9.0 at 25 °C.

In summary, the substance has a hydrolysis half-life of about 4 days under neutral pH conditions at 25 °C and a significantly longer half-life under acidic or alkaline pH conditions.
