Description of key information

HA-QSAR predictions are available to assess the aquatic toxicity of the registered substance, Reinblau BLW (CAS No 32724-62-2), in acute and/or chronic exposure.


A category approach has been defined (see attached justification document in section 13). The category consists of substances all having the diamino-anthraquinone structure as a common moiety which is linked to phenyl groups via the amino groups. Differences within the category are described by various alkyl groups bound to the phenyl groups. All substances included in the category approach are poorly water soluble with high adsorption coefficient (log Koc>6), high partition coefficient (logKow>8) and not readily biodegradable. These properties lead to a reduced bioavailability for organisms.


Within the category, no toxic effects were observed up to the highest attainable limit of solubility of the substances in experimental acute studies for the three trophic levels (fish, aquatic invertebrates and algae). However, due to the poorly water solubility and the very adsorptive behavior of the substances, the available experimental short-term toxicity studies cannot be used to establish a trend for the aquatic toxicity of the substances. These short-term tests do not give a true measure of toxicity for this type of substance and the long-term toxicity tests are required. To fulfill these chronic endpoints, HA-QSAR predictions, experimental studies and testing plans for long-term exposure are available/requested. Based on available information at the time being, no toxic effects were estimated or observed up to the highest attainable limit of solubility of the registered substance, Reinblau BLW, and all members of the category approach, in chronic exposure too. 


The registered substance, Reinblau BLW, is not expected to be hazardous to pelagic organisms.


Regarding toxicity to microorganisms, based on the experimental study performed on the source substance, Reinblau RLW (CAS No 41611-76-1), a member of the category, no effect on the respiration rate of activated sewage sludge are expected at the limit of water solubility of the substance, neither at concentrations in excess (no effects up to 1000 mg/L). 


Two news studies are planned on the registered substance (OECD TG 201 and 209 studies). The dossier will be updated with the new studies as soon at it become available.
