Biodegradation in water and sediment: simulation tests
Administrative data
- Endpoint:
- biodegradation in water: sewage treatment simulation testing
- Type of information:
- experimental study
- Adequacy of study:
- disregarded due to major methodological deficiencies
- Study period:
- 1986-01-02 to 1986-02-07
- Reliability:
- 3 (not reliable)
- Rationale for reliability incl. deficiencies:
- other: see 'Remark'
- Remarks:
- Non GLP-study. Deviations to the updated OECD Guideline (adopted 2001): A 0.45 µm filter was used. DOC measurement was solely used for monitoring the elimination of the nutrient solution. The elimination of test item was measured by specific analysis. Test item was not dissolved in the nutrient solution.

Study design
- Oxygen conditions:
- aerobic
- Inoculum or test system:
- sewage, domestic, non-adapted
- Details on inoculum:
- - Source of inoculum/activated sludge:
Activated sludge produced in the test apparatus from a mixed inoculum obtained from:
- Bacteria collected from the secondary effluent of the sewage treatment plant of CH-4153 Reinach.
- Water from the Rhine collected in CH-4000 Basle.
- A suspension of garden soil collected in CH-4000 Basle. - Duration of test (contact time):
- 36 d
Initial test substance concentration
- Initial conc.:
- 29.72 mg/L
- Based on:
- test mat.
- Parameter followed for biodegradation estimation:
- test mat. analysis
- Details on study design:
- TEST CONDITIONS
- Volume of test solution/treatment:
Test item was dissolved in dichloromethane to produce a stock solution of 3.57 g/L . The concentration was confirmed analytically and was within +/- 2.3 % of the nominal value.
- Composition of medium: in accordance to the guildeline
- Test temperature: The room temperature was kept at 23°C +/- 2 The temperature in the aeration vessel was maintained at 2°C below the room temperature.
- Other: Nutrient solution from the same batch as the blank unit. The test substance was pumped directly into the aeration vessel at 8.33 ml/h to produce a nominal concentration of 29.72 mg test item/L in the aeration vessel. Due to the turbulance produced by the air blown into the aeration vessel it was assumed that the test item was homogenuously distributed.
TEST SYSTEM
- Culturing apparatus:
All glass Husman - apparatus with a 3 L aeration vessel with 2 containers of 30 Lcapacity for receiving the nutrient solution and the effluent.
- Method used to create aerobic conditions: aeration vessel
- Measuring equipment:
Carbon analyzer for the determination of the DOC. HPLC-analyzer for the specific detection of the test substance.
The oxygen values in mg/1 in the aeration vessels of the test and blank unit were measured with a WTW OXI 91 oxygen meter with EO 90 electrode. The pH was measured in the aeration vessels of the test and blank units with a Knick PORTATEST 901 pH - meter and a EA 120 electrode.
CONTROL AND BLANK SYSTEM
- Inoculum blank: nutrient solution containing an average of 111.5 mg DOC/L. ( max: 120.1 mg DOC/L min : 105.6 mg DOC/L)
