The study was conducted as per the method outlined in the publication - Alsop, G.M.; Waggy, G.T.; Conway, R.A., Bacterial Growth Inhibition Test, Journal Water Pollution Control Federation, Volume 52, No. 10, October 1980. The test material n-pentyl propionate was evaluated at selected conentrations in a mixture containing buffer, nutrients, growth substrate and microorganisms. The mixture of 1 ml of a suspension of seed microorganisms, 20 ml of dilution water, 4 ml of stock buffer solution, 10 ml of yeast extract/sodium acetate solution and 4 ml of aqueous solution of n-pentyl propionate were incubated in an 8-ounce, narrow neck, round bottle for 16 hours on a platform shaker at ambient temperature (22 ± 2 °C). Seeded control bottles were used to measure growth or turbidity generated during the 16 hours without the test material. The bottles were stoppered with cotton plugs during shaking to avoid contamination.
The degree of inhibition was assessed from measuring (optical density at 530 nm) the turbidity levels of the test material at various concentrations. The measured optical density values were calculated as a percentage of the seeded growth control system. The percentage of control values were then plotted against the log of test sample concentration. The test concentration corresponding to 50 percent of the control was termed as 50 percent inhibition concentration (IC50).
