- Concentrations: (Nominal) 0; 125; 250; 500; 1,000 and 2,000 mg/L
- Sampling method: Samples were collected at test start and at the end of the test for chemical analysis of the test product. Duplicate samples of 12 mL were collected in 20-mL plastic vials for later analysis. Samples from the highest test concentration 2,000 mg/l were analysed as no effects were seen in any of the concentrations. Although duplicate samples were collected, only one of these samples was analysed. The spare samples were discarded after sponsor approved the report.

A stock solution of 2,000 mg/L was prepared by weighing out 2,000 mg of the test product and dissolving it in 1 L of test medium. pH in the stock solutions was 9.2 and was therefore adjusted to 7.9 by adding 0.1 M and 1 M HCl. The test solutions were prepared by diluting the appropriate amounts of stock sollution in test medium.
The test product was tested at the following nominal concentrations: 0 (control); 125; 250; 500; 1,000 and 2,000 mg/L.

- Source: A strain of freshwater crustacean Daphnia magna (Straus) Cladocera, Crustacea originally isolated from Langedam (Birkerød, Denmark), have been cultured at DHI since 1979. Animals from this culture were used in the study.

The chemical analysis of IBP1-Na conducted by Cheminova A/S showed a measured IBP1-Na concentration of 103-106% of the estimated nominal concentration of IBP1-Na. As the analysed concentration was maintained within ± 20% of the nominal concentration, the test product is assumed to be stable in all test concentrations.

< 10 of control animals were immobilized. Dissolved ozygen saturation at the end was ≥ 98% in both tests and controls. EC50(24h) = 1.6 mg/L for control substance (K2Cr2O7, CAS no 7778-50-9) which in the specified range (0.6-2.1 mg/L)
Conclusions:
EC50(48h) of test item is exceeds 1020 mg/L for Daphnia magna.
Remark: the test solution contains NaOH due to the production method.
Executive summary:

The short term toxicity to aquatic invertebrates was done according to the OECD guideline202 and ISO 6341 under GLP compliance. The test was done on Daphnia, they were exposed to the test item for 48hours. As only 5% of


immobilization was observed at 1000 mg/L and no significant effect was showed at the tested concentrations of the sample. Thus EC50(48h) value of the test item is determined >1020 mg/L.
