PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: the test substance solution was prepared by mixing 798.8 mg of test substance with 801.16 mg Cremophor EL and adding this micture quantatively to 8 litres of ISO-medium providing a nominal concentration of 100 mg/L. This dispersion was stirred for ca. 66 hours before the start of the exposure. The final solution was a turbid, white suspension.

EFFECT PARAMETERS MEASURED: mortality after 2, 24, 48, 72 and 96 hours. In addition, Dissolved oxygen content and pH were measured daily in all vessels, beginning at day 0. Temperature of the medium was measured daily in one control vessel, beginning at day 0. And fish length and weight was measured in 10 fish of the batch used for the test, prior to the start of the test.

Conclusions:
Under the conditions of this study, the 96h-LC50 for carp exposed to Alcamizer 5 was above the solubility limit of the test substance in test medium. Even though analysis of the test solution showed that no levels of Al or Mg higher than those in the controls were found, the turbid, white suspension proved that the fish were exposed to the test substance.
Executive summary:

Quantative analyis was performed on Al and Mg in samples taken from 0 and 100 mg/L and the Cremophor control to determine the test concentrations. Analysis showed that no levels of Al or Mg higher than those in the controls were found in the 100 mg/L solution. Measured concentrations of test substance, based on Al were calculated to be < 1.52 mg/L in both controls and 100 mg/L solution. Water solubility was determined to be < 2.4 mg/L in an earlier study. The 100 mg/L solution was a turbid, white suspension which proved that the fish were exposed to the test substance.
