- PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: Preparation of test solutions started with loading rates individually prepared in the range of 1.0 and 100 mg/L. A two-day period of magnetic stirring was applied to ensure maximum dissolution of the test item in test medium. Thereafter, the aqueous Water Accommodated Fractions (WAF’s) were collected by filtration through a 0.45 µm membrane filter (RC55, Whatman) and used as test concentrations. In the final test, the pH of the WAF prepared at a loading rate of 1.0 mg was adjusted from 8.0 to 8.5 with 1N NaOH (Merck, Darmstadt, Germany). The pH of the WAFs prepared at loading rates in the range of 3.2 to 100 mg/L were adjusted from 8.8-11.4 to 8.2-8.5 with 0.1 and 1.0 N HCL (Merck, Darmstadt, Germany).

- A dose-related increase of algal growth inhibition was observed with increasing loading rate, whereby the effects observed at loading rates of 10 and 32 mg/L were comparable. The inhibition of both growth rate and yield was statistically significant at all loading rates. The inhibition of growth rate observed at a loading rate of 1.0 mg/L was below 10% and thus considered to be biologically not relevant.

- Executive summary:
A growth inbition test on Pseudokirchneriella subcapitata was performed with the test item according to the OECD Guideline 201 (GLP study). As the test item was not completely soluble in test medium at the loading rates initially prepared, Water Accomodated Fractions were individually prepared at loading rates ranging between 1.0 and 100 mg/L and used as test concentrations. A final test was performed based on the results of a preceding combined limit/range-finding test. Six exponentially growing algal cultures of P. subcapitata were exposed to an untreated control, whereas three replicates per group were exposed to WAFs prepared at loading rates of 1.0, 3.2, 10, 32 and 100 mg test item per litre. The initial algal cell density was 10^4 cells/mL. The total exposure period was 72 hours. The algal cultures were assessed for the growth by visual cell count at 24, 48 and 72 h. No analysis of the test media was conducted because the development of an analytical method was not feasible. The study met all acceptability criteria and was considered valid. A concentration-effect relationship was observed and statistically analysed to obtain effect concentrations. The test item reduced growth rate and inhibited the yield of this fresh water algae species significantly at WAF prepared at loading rates of 1.0 mg/L and higher. The growth rate inhibition effect concentrations with their respective 95% lower and upper confidence limits for the inhibition of growth rate were ErL10 = 9.4 (8.6 - 10) mg/L, ErL20 = 15 (14 -16), ErL50 = 36 (35 - 38) mg/L. The yield inhibition effect concentrations with their respective 95% lower and upper confidence limits for the yield were EyL10 = 0.44 (0.39 - 0.50) mg/L, EyL20 = 0.90 (0.82 - 0.98) mg/L, EyL50 = 3.5 (3.4 - 3.7) mg/L. The 72h-NOELR for both growth rate and yield was 1.0 mg/L based on biological relevance.
