Duplicate samples were taken from the test media of all test concentrations at the start of the test (without algae) and at the end of the test (containing algae). For the 72-hour stability samples, the contents of the respective replicates were combined prior to sampling.
All samples were stored frozen (at about -20 ± 5 °C) immediately after sampling.

PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: An aqueous solution with a nominal content of 80 % was prepared prior to the test. The test substance content of this working solution was analytically verified by HPLC to be 77.9 % (water-free).
A stock solution of the nominal concentration of 52 mg/L was prepared by completely diluting 67 mg of the working solution (corresponding to 52.2 mg water-free test substance) in 1000 mL of test water by intensive stirring at room temperature for 30 minutes in the dark.
The stock solution was serially diluted with test water to prepare the test media of the two highest test concentrations. The test media of the lower test item concentrations were prepared in a series of further dilution steps.
The test media were prepared just before the start of the test.

- Age of inoculum (at test initiation): An inoculum culture was set up four days before the start of the exposure. The algae were cultivated under the test conditions. The inoculum culture was diluted threefold one day before the start of the test to ensure that the algae were in the exponential growth phase when used to inoculate the test solutions.

The temperature in the incubator was monitored and recorded continuously. The appearance of the test media was also visually controlled and recorded daily.

The light intensity over the incubation area was within a ±15 %-deviation from the average light intensity as recommended by the guideline.
