Description of key information

Methyl gentisate is the methyl ester of the naturally occurring gentisic acid. Gentisic acid contains two phenolic hydroxy groups that may be conjugated by phase-II xenobiotic metabolising enzymes such as sulfotransferases or UDP-glucuronosyl transferases.

Methyl gentisate was found to permeate the skin, and considering the measured log POW of 1.8, it is also expected to be taken up via the oral route or if inhaled as a dust. Both the log POW of less than 3 and the presence of metabolisable functional groups suggest the substance has no potential for bioaccumulation.

In 2006, methyl gentisate was described as a relatively new skinlightening agent which is structurally similar to hydroquinone (Gallo R.and Baldary M. Contact Dermatitis 2006: 54 220 - 221).

Methyl gentisate in vitro inhibits pigmentation in melanocytes, inhibits tyrosinase selectivity, and is less cytotoxic than hydroquinone.
