Details on test organisms:
Test system
The study was performed with earthworms Eisenia foetida bred by RCC under standardised conditions (as per OECD 207). Only adult worms (older than two month) with clitellum and body weight of 300-600 mg were used; they could acclimate to artificial soil for 24 h before test start.

Total exposure duration:
14 d
Details on test conditions:
Test conditions
At test start, ten worms were placed on the surface of the artificial soil of each test vial (four replicates per concentration/control). The test vials were exposed in an air conditioned room at 20 ± 2 °C. The temperature in the test room reached at times 25 °C, but this could have, if at all, only little influence on the mortality of the worms. The continuous illumination was measured to be 330-650 lux during the entire study. The vials were controlled at days 7 and 14 respectively.
Nominal and measured concentrations:
0, 0.1, 1.0, 10.0, 100 and 1000.0 mg/kg

pH and water content of the artificial soil

During the entire study, the pH of the artificial soil was in a range of 5.6 to 5.9 and did not deviate from the values allowed by the guidelines (6.0 ± 0.5). The water capacity of the artificial soil was determined to be 55% of the dry weight at the begin of the study. The water content was 35.3 to 35.7 % (dry weight) at start and in a range from 33.2 to 36.3% at the end of the study (see table 2). This does not differ significantly from the value given by the guidelines (ca. 35 %).

Mortality and body weights of E. foetida

The mortality of E. foetida increased with increasing concentration of DMPPA_701-402-5 in the artificial soil. This tendency could be seen already after 7 days of experimental time and was confirmed at the end of the study. So within 14 days 12 % of the worms died in the vials containing 0.1 mg/kg and 35 % in those vials containing 1000 mg/kg of the test substance. Nevertheless, the mortality of E. foetida even in the vials with the highest concentration of

DMPPA_701-402-5 (as well as in all other test substance treated vials) was not significantly higher than in the water treated controls (Kruskal-Wallis - test, p < 0.05 and Dunn - test, p < 0.05). Taking the results of the pre-test and main test of this study into consideration it is obvious that DMPPA_701-402-5

affects E. foetida, although for the concentrations tested here no acute toxicity could be proved. According to this results it did not make sense to calculate the LC50 of the test substance, which clearly must be more than 1000 mg/kg of DMPPA_701-402-5 in artificial soil. In all test concentrations, the body weights of the surviving worms decreased lightly due to the starvation of the animals within the test period. No behavioural abnormalities could be observed.

Validity criteria fulfilled:
yes
Conclusions:
The LC50 (14 days) was >1000 mg/kg soil.
Executive summary:

In a 14 days lasting study the mortality of Eisenia foetida under the impact of DMPPA_701-402-5 was determined according to OECD 207 guideline and in compliance with Good Laboratory Practice regulations. The concentrations used were 0, 0.1, 1.0, 10.0, 100 and 1000.0 mg/kg dry weight. 12 % of the worms died in the vials containing 0.1 mg/kg and 35 % in those vials containing 1000 mg/kg of the test substance. Hence, the LC50 (14 days) was >1000 mg/kg soil.
