There are no studies available, in which the ready biodegradability of the target substance N-(1-oxotetradecyl)sarcosine (CAS 52558-73-3) was assessed. Therefore, read-across to the two structurally and chemically closely related source substances Sodium N-methyl-N-(1-oxotetradecyl)aminoacetate (CAS 30364-51-3) and N-methyl-N-[C18-(unsaturated)alkanoyl]glycine (EC 701-177-3) was performed in accordance with Regulation (EC) No 1907/2006 Annex XI, section 1.5 in order to fulfill the data requirements for this substance. Dissociation of the source substance Sodium N-methyl-N-(1-oxotetradecyl)aminoacetate (CAS 30364-51-3) (sodium salt) yields identical ionic species as dissociation of the target substance (acidic form) under environmental conditions. The source substance N-methyl-N-[C18-(unsaturated)alkanoyl]glycine (EC 701-177-3) is identical in structure as the target substance and only differs by a slightly longer carbon chain length (C18). Since the determining factor for biodegradability is carbon chain length, this source substance (acid form) is considered a worst-case approach based on its longer chain length compared to the target substance (C14), and was therefore selected as key study. Thus, the selected source substances are considered suitable representatives for the assessment of the biodegradability of the target substance. A detailed read-across justification in provided in IUCLID section 13.
The key study was performed with the structurally and chemically related source substance N-methyl-N-[C18-(unsaturated)alkanoyl]glycine (EC 701-177-3), according to OECD guideline 301 B and GLP. In this study, 20 mg/L test item (based on TOC) was inoculated with 25 mg/L domestic, non-adapted activated sludge for 28 d. After 28 d, the test item was degraded by 85.2% in terms of CO2 evolution. Thus, the source substance N-methyl-N-[C18-(unsaturated)alkanoyl]glycine (EC 701-177-3) is readily biodegradable according to guideline criteria.
Two GLP studies are available for the source substance Sodium N-methyl-N-(1-oxotetradecyl)aminoacetate (CAS 30364-51-3), conducted according to OECD 301 B and 310.
In the study according to OECD 301B, a concentration of 104.95 mg/L test item was inoculated with activated sludge for 28 d. After 28 d, a mean degradation of 83% was observed but the 10-day window was not met by only 1 d (60% on Day 14 instead of Day 13). In the study according to OECD 310, domestic activated sludge was used as inoculum with a test item concentration of 20 mg/L based on TIC. After 28 d, a mean biodegradation of 71% mean was recorded but the 10-day window was not achieved by 1 d (60% biodegradation on Day 13, instead of Day 12).
Based on the structural and chemical similarity of the target and source substances, the target substance is expected to exhibit a similar environmental fate profile as the selected source substances. Since the key study with the source substance N-methyl-N-[C18-(unsaturated)alkanoyl]glycine (EC 701-177-3) represents a worst-case scenario with regard to carbon chain lengths and the 10-d window in both supporting studies was only narrowly missed by 1 d, it is concluded that the target substance N-(1-oxotetradecyl)sarcosine (CAS 52558-73-3) is readily biodegradable according to OECD guideline criteria.
