Since no studies investigating the short-term toxicity of Fatty
acids, coco, decyl esters (CAS 93455-79-9) to fish are available, in
accordance to Regulation (EC) No 1907/2006 Annex XI, 1.5 a read-across
to the structurally related source substances Tetradecanoic acid,
tetradecyl ester (CAS 3234-85-3) and Fatty acids, C8-10, C12-18-alkyl
esters (CAS 95912-86-0) was conducted. The read-across substances are
representative to evaluate the short-term toxicity of Fatty acids, coco,
decyl esters (target substance) to fish.
The target substance is a UVCB substance specified by mainly C12 linear
fatty acids esterified with a linear alcohol of a chain length of C10.
The first source substanceTetradecanoic acid, tetradecyl esteris a
mono-constituent ester of a linear C14 fatty acid and a linear C14
alcohol. The second source substance Fatty acids, C8-10, C12-18-alkyl
esters is a UVCB substance predominantly consisting of esters with a
C8-C10 fatty acid and a C12-C14 alcohol moiety. Both substances have a
comparable similar structure and are suitable for read across to the
target substance.In this case of read-across, the best suited (highest
degree of structural similarity, nearest physico-chemical properties)
read-across substance was used for the assessment.
This read-across is justified in detail in the overall summary (IUCLID
Section 6.1) and within the analogue justification in IUCLID Section 13.
One GLP Guideline study according to OECD 203 is available investigating the short-term toxicity of the source substance Tetradecanoic acid, tetradecyl ester (CAS 3234-85-3) to freshwater fish (Teigeler, 2012). The zebra fish Danio rerio was exposed for 96 h to a limit Water Accommodated Fraction (WAF) concentration of nominal 100 mg/L.GC/EI-MS analysis resulted in a measured initial concentration of 0.189 µg/L which dropped below the detection limit of 0.1 µg/L at the end of exposure after 96 h. No mortality was observed resulting in a LL50 (96 h) of > 0.189 µg/L (measured initial concentration) and > 100 mg/L (nominal), respectively.
The study with the second source substance Fatty acids, C8-10, C12-18-alkyl esters was performed according to OECD 203 (GLP) investigating the toxicity of to fish (Salinas, 2013). Danio rerio was exposed to a nominal concentration of 100 mg/L for 96 h (Water Accommodated Fraction). No mortality was observed after 96 h of incubation resulting in a LL50 (96 h) > 100 mg/L (nominal) and a NOELR (96 h) ≥ 100 mg/L (nominal).
Based on the available results from structurally related source substances (in accordance to Regulation (EC) No 1907/2006 Annex XI, 1.5) which are characterized by a comparable structure, it can be concluded that Fatty acids, coco, decyl esters will not exhibit short-term effects to fish up to the limit of water solubility.
