Study design

Details on sampling:
Analysis of the Sample Solutions
The sample solutions were taken from the water bath at 24 and 120 hours and the pH of each solution recorded.
The concentration of test item in the standard, sample and matrix blank solutions was determined by liquid chromatography – mass spectroscopy (LC-MS) using the conditions detailed in the Analysis Section below.
Samples
An aliquot of each sample solution was diluted by a factor of 2 using tetrahydrofuran.
Standards
Calibration standard solutions of test item were prepared in THF:buffer (1:1 v/v) in the range of 0.01 to 2.5 mg/L with a duplicate standard prepared at a nominal concentration of 0.5 mg/L.
Matrix Blank
THF:buffer (1:1 v/v)

Further Testing
Results from the Preliminary Test indicated that additional testing was required.

Testing was performed in 10% pH 9 buffer at 37 °C over a period of 24 hours of incubation and analyzed directly from vials containing test solution in the heated sampling tray of the instrument.

Preparation of the Test Solutions
A stock solution of test item was prepared at a nominal concentration of 300 mg/L in tetrahydrofuran. An aliquot of the stock solution (1 mL) was added to 100 mL of a 10% solution of pH 9 buffer to give a nominal concentration of 3 mg/L of test item (matrix THF:10% buffer 1:99 v/v).
The test solution was transferred to low adsorption amber vials, placed in the autosampler set at a temperature of 40 °C and analysed at multiple time points over a period of approximately 21 hours.

Analysis of the Sample Solutions
The concentration of test item in the sample solutions was determined by liquid chromatography – mass spectroscopy (LC-MS).

Samples
Samples were injected directly from the incubated vials.
Standards
Calibration standard solutions of test item were prepared in THF:buffer (1:99 v/v) in the nominal concentration range of 0.1 to 5 mg/L with a duplicate standard prepared at a nominal concentration of 3 mg/L.
Matrix Blank
THF:buffer (1:99 v/v)
Buffers:
The test system consisted of sterile buffer solutions at pH’s 4.0, 7.0 and 9.0.

Details on test conditions:
Preliminary Test / Tier 1
Preparation of the Test Solutions
A stock solution of test item was prepared at a nominal concentration of 100 mg/L in tetrahydrofuran. An aliquot of the stock solution (10 mL) was added to 1 L of a 10% solution of each buffer to give a concentration of 1 mg/L of test item in each buffer. The test solutions were split into individual glass vessels for each time point. The solutions were shielded from light whilst being maintained at the test temperature.
Sample solutions at pH 4, 7 and 9 were maintained at 50.0 ± 0.5 °C for a period of 120 hours.

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Conclusions:
Determination of the rate constants and half-lives of the test item at pH 4, 7 and 9 were not possible due to surface effects of the test item.

Evaluation of the testing is summarized below:

The hydrolysis of the test item was evaluated at pH 4, 7 and 9 at 50 °C for 120 hours and at pH 9 at 37 °C for approximately 21 hours. Test item concentrations for the C16-C18 alkyl amine components were observed to decrease. This was considered to be due to surface effects rather than hydrolysis, since there are no hydrolyzable groups present and fatty amines are known to strongly adhere to surfaces by chemical or physical processes. Test item concentrations for the dibuylthiophosphate component were not observed to change significantly over the course of testing the and therefore this component was considered to be stable at pH 4, 7 and 9.
Executive summary:

The determination was carried out using a procedure designed to be compatible with Method C7 Abiotic Degradation, Hydrolysis as a Function of pH of Commission Regulation (EC) No 440/2008 of 30 May 2008 and Method 111 of the OECD Guidelines for Testing of Chemicals, 13 April 2004.
