The part of a chemical substance that is able to be oxidized is determined. The sample is mixed with potassium dichromate (serving as oxidizing agent) and stirred for 2 hours in a sulfuric acid solution at elevated temperature. Subsequently, the excess of dichromate is determined quantitatively by titration with ammonium iron(II)-sulfate solution.

In a key study the test substance FAT 65004 showed 0 % biodegradation under MITI - Test, OECD 302 C.

In a supporting study was performed to determine the biochemical oxygen demand of FAT 65004 by measuring the dissolved oxygen concentration before and after 5 days incubation at 20 °C. Based on the findings from the study, the BOD5 of FAT 65004 is 35 mg O2/g test material, and COD was noted to be 180 mg O2/g test material. The relevant BOD5/COD quotient is <0.5, therefore the substance is not indicative of rapid degradation.

Taking into consideration the data from key study and supporting studies, the test substance FAT 65004 is considered to be not biodegradable.

By the nature of their design and use, optical brighteners are not intended to be readily biodegradable as this would assist in the rapid destruction of the chemical, rendering it unfit for purpose. As such, it is accepted that such substances are not readily biodegradable under relevant environmental conditions. A published study (Pagga & Brown, 1986) describes the results of the testing of 87 dyestuffs in short-term aerobic biodegradation tests. The authors of this publication concluded that dyestuffs are very unlikely to show any significant biodegradation in such tests and that there seems little point in carrying out such test procedures’ on dyestuffs
