All concentration levels and the control were analytically verified via LC-MS/MS after 0 hours (start of the exposure) and 72 hours (end of the exposure).
For the test item analysis at the beginning of the exposure, separate replicates for each test item concentration and control were prepared with algae. For the test item analysis after 72 hours, samples were taken from the test replicates.

Details on test solutions:
The test item is known to degrade quickly (half-life < 1 hour) by hydrolysis. Therefore, the hydrolysis product was tested as recommended in OECD Guidance Document No. 23 (2000). For this purpose, a stock solution of 100 mg/L of the test item was prepared with dilution water and stirred for 24 hours with 1100 rpm at room temperature before the start of the exposure phase to ensure complete hydrolysis of the test item. Thereafter, undissolved particles were removed by membrane filtration (membrane filter 0.45 µm, RC, MACHEREY-NAGEL). During filtration the filter was always kept covered. The filter was saturated in order to avoid adsorption during the filtration. The first 25 mL filtrate were discarded. The filtration was interrupted for ca. 15 minutes to allow adsorption and saturation of the filter material with dissolved test item. Thereafter, the filtration was continued. The next 25 mL were discarded. The following filtrate (visually clear and colourless) was used in the test. Further dilution levels were prepared from the saturated solution

Cultivation at test facility: Fresh stocks are prepared every month on Z-Agar. Light intensity amounted to 35-70 µE·m-2·s-1 for 24 hours per day.

A four days old preculture, prepared in dilution water, was used as inoculum.

Nominal and measured concentrations:
The measured concentrations of the test item at the start of the exposure reflect the dilution factor of 2.5. At the end of the exposure, the measured concentrations of the test item were within the range of 86 to 95% of the initially measured concentrations.
5 concentrations were tested in a geometrical series with a dilution factor of 2.5: 2.56 - 6.40 - 16.0 - 40.0 - 100 % of the saturated solution. These dilution levels are corresponding to geometric mean measured test item concentrations of 0.503 - 1.21 - 3.19 - 7.92 - 19.2 mg/L. The concentrations were based on the results of a preliminary range finding test (non-GLP).

Details on test conditions:
The cell density was measured daily via Chlorophyll a-fluorescence, excitation at 436 nm, emission at 685 nm. Dilution water was used as a background signal. No self-fluorescence was found in the definitive test at the saturated solution.

Conclusions:
In this study, teh test item was found to inhibit the growth of the freshwater green alga Pseudokirchneriella subcapitata after 72 hours with the following effect values (based on geometric mean measured test item concentrations): The EC50-values with 95 % confidence intervals for inhibition of growth rate (ErC50) and yield (EyC50) after 72 hours were > 19.2 and 12.4 (9.41 – 16.8) mg/L, respectively. The NOEC-values for both inhibition of growth rate and yield after 72 hours were 3.19 mg/L.
Executive summary:

The toxicity of the test item to the unicellular freshwater green alga Pseudokirchneriella subcapitata was determined according to the principles of OECD 201 and Council Regulation (EC) No. 761/2009/Method C.3.

The study was conducted under static conditions with an initial cell density of 6305 cells/mL. A saturated solution of 100 mg/L was prepared and five dilution levels were tested in a geometrical series with a dilution factor of 2.5:2.56 - 6.40 - 16.0 - 40.0 - 100% of the saturated solution. These dilution levels are corresponding to geometric mean measured test item concentrations of 0.503 - 1.21 - 3.19 - 7.92 - 19.2 mg/L.

Three replicates were tested for each test item concentration and six replicates for the control. The environmental conditions were within the acceptable limits.The test media were clear throughout the test period.
