Pharmacopoeia, Fourth

Edition, 2014

Principle of method

Pour Density:It is the ratio of known weight of test item and its volume in graduated

glass cylinder.

Tap Density: It is the

difference between the

volume of test item

measured before & after tapping in the graduated glass cylinder.

Conclusions:
The density of test item [8-[(4-amino-3-nitrophenyl)azo]-7-hydroxy-2- naphthyl]trimethylammonium chloride (CAS No. 68391-32-2)was determined following OECD Guideline 109 and according to the Test methods: Pour density and tapped density of powders” recommended in The International Pharmacopoeia, Fourth Edition, 2014 using bulk density apparatus . The mean pour density and tapped density of test item was determined to be 0.5570 g/cm³ and 0.7162 g/cm³at 28.4°C and 972.9 hPa.
Executive summary:

Additional information

Other supporting study from review article also indicates that the density of chemical (8-((4-Amino-3-nitrophenyl)azo)-7-hydroxy-2-naphthyl)trimethylamm- onium chloride was determined to be 1.7 g/cm3.

As the data from lab has K1 reliability and the test was performed using OECD guidelines which has the value of density as 0.5570 g/cm3 (pour density) and 0.7162 g/cm3 (tapped density) to,thus considered as key study for the chemical safety assessment
