L-Threonine did not show mutagenicity in several in vitro studies. It was negative in the Ames test (4 studies), in a mammalian cell gene mutation assay and in in vitro studies analyzing clastogenic effects (3 studies).

Further, L-threonine showed no clastogenicity in two in vivo micronucleus tests according to OECD guideline 474.

These results were foreseeable as L-threonine is a naturally occurring essential amino acid which is present in significant amounts in human body fluids (e g human blood plasma > 100 µM - LA Cynober 2002) as well as in human cells. It is a basic metabolite and building block of all living organisms and therefore a genotoxic/mutagenic potential could be excluded.
