Reference
Endpoint:
short-term toxicity to aquatic invertebrates
Type of information:
experimental study
Adequacy of study:
key study
Reliability:
1 (reliable without restriction)
Rationale for reliability incl. deficiencies:
other: GLP - Guideline study
Qualifier:
according to guideline
Guideline:
OECD Guideline 202 (Daphnia sp. Acute Immobilisation Test)
GLP compliance:
yes
Analytical monitoring:
yes
Details on sampling:
At the start and end of exposure, 0.75 mL of test liquid was sampled from 1 test container of each test group, and after acetonitrile was equally added and stirred into this, it was analyzed using HPLC. The test material concentration of each test liquid was quantified from the ratio with the standard solution peak area to find the geometric mean.
Vehicle:
no
Test organisms (species):
Daphnia magna
Details on test organisms:
TEST ORGANISM
- Japanese name: Oomijinko
- Source: Environment Agency, National Institute for Environmental Studies
- Age at study initiation: Male juvenile (born within 24 hours)
- Feeding during test: not fed during test

ACCLIMATION
- Acclimation period: rearing in conditioned water as used for test, prepared according to OECD Guideline 211; 1 organism/80 mL (25 organisms/2 L) or less
- Acclimation conditions: same
- Type and amount of food: Chlorella vulgaris (monocellular green algae)
- Feeding frequency: 0.2 mg C/organism/day
- Health during acclimation: Parent mortality rate 2 weeks prior to start of exposure: < 5%
Test type:
static

no
Total exposure duration:
48 h
Hardness:
Approx. 250 mg/L (CaCO3 conversion)
Test temperature:
20 +/- 1°C
pH:
The test liquid pH was not adjusted. During study: see table 3 . For test substance concentration 100 mg/L pH was only 5.1 and therewith outside the suitable range. This was thought to be due to the test material because the pH was low for a high concentration group at preparation.

Description of key information

The EC50 (48h) is 73.1 mg/L (geometric mean of measured concentrations).

Key value for chemical safety assessment

Fresh water invertebrates

Fresh water invertebrates
Effect concentration:
73.1 mg/L

Additional information

During the acute static immobilisation test with Daphnia magna according to OECD 202 90% of the daphnids were immobilised at the highest tested concentration of 100 mg/L (nominal). The EC50 was determined to be 73.1 mg/L (geometric mean of measured concentrations). Thus, the substance shows a toxicity to Daphnia magna under conditions tested.
