Endpoint:
short-term toxicity to aquatic invertebrates
Type of information:
experimental study

1 (reliable without restriction)
Rationale for reliability incl. deficiencies:
guideline study
Qualifier:
according to guideline
Guideline:
OECD Guideline 202 (Daphnia sp. Acute Immobilisation Test)

no
GLP compliance:
yes (incl. QA statement)
Analytical monitoring:
yes
Details on test solutions:
PREPARATION AND APPLICATION OF TEST SOLUTION
- Method: Water Accommodated Fractions (WAFs) were prepared by weighing portions of the test substance on small plastics trays and subsequently transferred from the trays to 1 liter of distilled water.
Test organisms (species):
Daphnia magna
Details on test organisms:
TEST ORGANISM
- Common name: Daphnia magna, Straus
- Source: own rearing, the original source Water Research Institute, Bratislava, Slovak Republic
- Age: < 24 h
- Feeding during test: no
Test type:
static
Water media type:
freshwater
Total exposure duration:
48 h
Hardness:
159,86 mg/L (CaCO3)
Test temperature:
19.0 °C - 20.9 °C
pH:
7.65 - 8.39
Dissolved oxygen:
7.23 - 7.91
Nominal and measured concentrations:
Nominal test concentrations: 20 mg/L, 40 mg/L, 60 mg/L, 80 mg/L, 100 mg/L (WAF)
Analysis of the WAF loading of the test substance at the start and end of the definitive test (100 mg/L WAF loading of the test substance) was sufficient because of the exposure WAF loading varied less than 20 % from nominal values during the test (+16.75 %).

EFFECT PARAMETERS MEASURED:
Immobilisation, measured daily

RANGE-FINDING STUDY
- Results used to determine the conditions for the definitive study: The limit test indicated that 48 h EC50 of the test substance in the test medium was lower than 100 mg/L (48 h immobilization 80 %).

Validity criteria fulfilled:
yes
Conclusions:
In an immobilisation test on Daphnia magna according to OECD 202, the 48h- EC0 was determined to 69.17 mg/L (WAF).
Executive summary:

The toxicity of MC A45A to Daphnia magna was determined in an immobilisation test according to OECD 202. Due to the limited water solubility of the test item, the test was therefore performed on water accommodated fractions (WAF).

Analysis of the test item performed during a limit test revealed that WAF loading varied less than 20% from nominal values.

The results of testing reference substance are consistent with literature data.
