A reliable study is available for skin and eye irritation in rabbits (1975): DOTG is not irritating for skin but slightly irritating for eyes in rabbit.

DOTG is brought on the intact or abraded skin under a surgical patch measuring 1 inch x 1 inch. The patches are fixed to the application site by means of adhesive tape and the entire trunk of the rabbits is wrapped with an impervious material to maintain the test patch in position.
Twenty four hours prior to applying the materials, the hair is removed from the backs of the animals with an electric clipper in such a way as to avoid abrasions.

Twenty four hours after DOTG had been brought into the eyes, slight corneal damage was observed in three out of six rabbits, slight iritis in four out od six rabbits and slight or moderate conjunctivitis in all test rabbits. The eye lesions recovered gradually but incompletely, so what at the seventh day reading slight conjunctivitis was still observed in three out of 6 rabbits.

DOTG caused slight corneal damage in three out of 6 rabbits, slight iritis in four out of 6 rabbits, and slight or moderate conjunctivitis in all tested rabbits. In conclusion, DOTG is slightly irritant for eyes.

Justification for classification or non-classification

Based on the available data, 1,3-di-o-tolylguanidine should not be classified as irritating according to the Regulation EC N°1272/2008.
