HPLC method

Details on study design: HPLC method:
EQUIPMENT
- Apparatus: HPLC
- Type: Waters 616 with autosampler Waters 717 plus
- Type, material and dimension of analytical (guard) column: YMC-Pack Cyano, cyanopropyl solid phase, 250 x 4.0 mm, YMC Europe GmbH
- Detection system: 2996 Diode Array Detector (DAD), Waters, for the reference items and IR detector HP 1047 A, Hewlett Packard, for the test item

MOBILE PHASES
- Type: 70:30 (% v/v) acetonritrile : HPLC water
- Experiments with additives carried out on separate columns: no
- Solutes for dissolving test and reference substances: acteonitril/water for test substance, methanol for reference substances

DETERMINATION OF DEAD TIME
- Method: by inert substances which are not retained by the column: NaNO3

REFERENCE SUBSTANCES
- Identity:
Acetanilide (CAS 103-84-4), Atrazine (CAS 1912-24-9), Monuron (CAS 150-68-5), 2,5-Dichloroaniline (CAS 95-82-9), Benzoic acid phenylester (CAS 93-99-2), Fenthion (CAS 55-38-9), Phenanthrene (CAS 85-01-8), 4,4¿-DDT (CAS 50-29-3)

DETERMINATION OF RETENTION TIMES
- Quantity of test substance introduced in the column: 100 µl of 400 mg/l solution
- Quantity of reference substances: 100 µl of 100 mg/l solution
- Intervals of calibration: each reference item was analysed separately before and after analysis of the test item

REPETITIONS
- Number of determinations: 3

EVALUATION
- Calculation of capacity factors k': k'= (retention time tR - dead time t0)/dead time t0
- Determination of the log Koc value: By means of the adsorption coefficient values (Koc) and the corresponding capacity factors of the reference items a calibration calibration graph of log k` vs. log Koc,soil and log k ' versus logKoc, sewage sludge was plotted. The Koc values of the test item was then calculated based on the calibration curves.

log Koc, sewage sludge = 9.79, Koc, sewage sludge=6.2x10exp9.

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Conclusions:
For 2-Octyldodecanol for soil the log Koc was determined to be 8.92 with a corresponding KOC value of 8.4x10exp8. For sewage sludge the log Koc was determined to be 9.79 with a corresponding Koc of 6.2x10exp9.
Executive summary:

At test conditions the retention time of the test substance 2 -Octyldodecan-1 -ol was found to be higher as the retention time of the reference item 4,4-DDT (highest Koc value in the set). Therefore the log Koc values were extrapolated.

For 2-Octyldodecanol for soil the log Koc was determined to be 8.92 with a corresponding KOC value of 8.4x10exp8. For sewage sludge the log Koc was determined to be 9.79 with a corresponding Koc of 6.2x10exp9.
