Materials and methods

Test guideline
Qualifier:
equivalent or similar to guideline
Guideline:
OECD Guideline 211 (Daphnia magna Reproduction Test)
Deviations:
not applicable
Remarks:
QSAR model adapted specifically to test guideline: OECD 211.
Principles of method if other than guideline:
The chronic toxicity of the daphnids was determined using a validated QSAR model for the Mechanism of Action (MechoA) in question (MechoA 1.1, i.e. non-polar narcosis) (3). The QSAR is based on validated data for a training set of 30 chemicals derived from 21-day test on daphnids, for which the concentrations of the test item had been determined by chemical analyses over the test period.

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Conclusions:
The NOEC 21d predicted with this QSAR calculation, on the basis of a water solubility predicted to be 7.02 mg/L, is 0.072 mg/L.

However, in water solubility assay performed otherwise, poor water solubility combined with high volatility made impossible to solubilize tetradecafluorohexane at higher concentration than 0.02 mg/L after more than 24 hours. Moreover, the solubility assay was performed in closed vessel to minimize volatization, with continuous stirring, and the concentrations measured were probably far higher than what could be expected in the environment or in a long term toxicity assay on daphnia.

Therefore, it can reasonably be concluded that the 21d NOEC predicted is above the limit of solubility of tetradecafluorohexane.
