Skin irritation:

Skin irritation was analyzed according to OECD 404. After 3 min of exposure 2-ethylhexylamine produced dermal corrosion at all sites. The dermal response was initially characterized by necrosis at three minutes and progressed to eschar (scab) at 24 hours post-dose. The eschar persisted through the remainder of the seven-day study. During the test period, a very slight to moderate edema, which diminished by the end the study, was noted (Hoechst AG 1988, reliability score: 1). In a primary skin irritation study with undiluted 2-ethylhexylamine exposure times of 1, 5 and 15 min caused irreversible leather-like necrosis or hard necrosis, both defined by complete destruction of the skin tissue involving all individual layers of the skin (BASF AG 1968; reliability score 2). In 24 h vesicant test the undiluted compound produced necrosis on 4 of 5 rabbits and the 4 h rabbit belly irritation test also indicated the production of necroses of the skin (Smyth et al. 1949; reliability score: 2).

 

Eye irritation:

In a primary eye irritation study in rabbits, 50 µL of the test substance were applied to the conjunctival sac of one eye in 2 animals and caused irreversible corrosive effects. After 8 days severe chemical burnings of eyelids and eye ball, iritis, crust formation on eye lids, secretion of pus, loss of bulbus was observed (BASF AG 1968; reliability score 2). A publication confirmed these severe corrosive effects (Smyth et al. 1949; reliability score: 2).

Justification for classification or non-classification

Classification, Labeling, and Packaging Regulation (EC) No. 1272/2008

The available experimental test data are reliable and suitable for classification purposes under Regulation 1272/2008. The substance is considered to be classified as skin corrosive cat. 1A (H314) and as causing severe eye damage cat. 1 (H318) under Regulation (EC) No 1272/2008, as amended for the tenth time in Regulation (EC) No 2017/776.
