key study
Study period:
2008-09-23 - 2009-01-29
Reliability:
1 (reliable without restriction)
Rationale for reliability incl. deficiencies:
other: The study was conducted according to an internationally accepted guideline. All study parameters are based on the specific guideline.
Qualifier:
according to
Guideline:
OECD Guideline 202 (Daphnia sp. Acute Immobilisation Test)
Deviations:
yes
Remarks:
A mobile phase consisting of 14 % methanol in water was used instead of 15 % methanol in water.
GLP compliance:
yes (incl. certificate)
Analytical monitoring:
yes
Details on sampling:
Analysis of test solutions for P 70/05/S-4 0 hours and 48 hours after test start resulted in concentrations ranging between 94 and 107 % of nominal. The test concentrations were found within ± 20 % of nominal throughout the test. Therefore, all results are based on the nominal concentration.
Vehicle:
yes
Details on test solutions:
The test item was dissolved directly in dilution water.
The test solutions were prepared in the following way:
- Weighing of 50.0 mg P 70/05/S-4
- Filling with dilution water up to 250 mL
(= stock solution A, containing 200 mg P 70/05/S-4/L)
The test solutions of the chosen concentration was prepared directly in the test vessels by dilution of the stock solution with dilution water immediately before the transfer of the Daphnia into the test vessels.
Test organisms (species):
Daphnia magna
Details on test organisms:
Holding and breeding in the test facility under standardised laboratory conditions,
Test animals derived from a healthy stock, not first brood progeny,
Age of the Daphnia used in the test: < 24 hours Holding conditions,
Breeding medium: M 4 medium (according to ELENDT, OECD 2002)
Breeding conditions: breeding was performed in glass beakers (50 mL) under the same conditions as in the test; test animals were separated from the adults by means of a pipette

Food: Alga Desmodesmus subspicatus, origin: Albrecht-v.-Haller-lnstitut für Pflanzenwissenschaften, Nikolausberger Weg 18, D-37073 Göttingen
Feeding: daily; Daphnia of different age were fed with a defined suspension of algae:≤ 1 day old: 0.5 mL/50 mL M 4, 3 to 10 days old: 1.0 mL/50 mL M 4, > 10 days old: 1.5 mL/50 mL M 4
Test type:
static
Water media type:
freshwater
Limit test:
no
Total exposure duration:
48 h
Hardness:
2 mmol/L CaC03 which corresponds to a hardness of 200 mg/L CaC03 (valid: > 140 and < 250 mg/L).
Test temperature:
19.5-20.1 °C
pH:
7.92
Dissolved oxygen:
8.58 mg/L
Salinity:
The sum of calcium and magnesium ions in the dilution water was 2.5 mmol/L. The proportion of Ca to Mg ions was 4:1 and of Na to K ions 10:1. The acid capacity Ka of the solution was 0.8 mmol/L.
Nominal and measured concentrations:
Analysis of test solutions for P 70/05/S-4 0 hours and 48 hours after test start resulted in concentrations ranging between 94 and 107 % of nominal. The test concentrations were found within ± 20 % of nominal throughout the test. Therefore, all results are based on the nominal concentration.
Details on test conditions:
Test vessel:glass beaker
Test volume:10 ml
Number of Daphnia/ test vessel (= replicate): 5
Number of test vessels/ concentration: 4 + 3 (1 for measuring, 1 for analysis, 1 retain specimen)
Number of daphnia/concentration: 20
Loading: 2 mL test solution per Daphnia

Test medium: dilution water: reconstituted water according to ISO 6341, prepared 1 day in advance, using deionised water conductivity of deionised water: ≤ 10 µ S/cm (measured 0.600 µS/cm)

Conclusions:
The short-term toxicity of N-[3(5)-Methyl-1 H-pyrazol-1 -yl-methyl]acetamide to aquatic invertebrates was determined according to OECD Guideline 202 (Daphnia sp. Acute Immobilisation). Under laboratory conditions no effects of the test item on immobilisation of daphnids were determined. The NOEC (no observed effect concentration) after 48 hours was determined to be 100 mg/L.
