Additional information

Since no experimental studies investigating the short-term toxicity of 1,3-Propnediol, 2,2-dimethyl-, C5-9 carboxylates (CAS 85711-80-4) to fish are available for this endpoint, in accordance to Regulation (EC) No 1907/2006 Annex XI, 1.5 a read across to the structurally related source substances Heptanoic acid, ester with 2,2-dimethyl-1,3-propanediol (CAS 68855-18-5) and 3,5,5-trimethylhexanoic acid mixed tetraesters with PE and valeric acid (CAS 131459-39-7) was conducted. This read-across is justified in detail in the overall summary (IUCLID section 6.1) and within the analogue justification in IUCLID Section 13.

The first study with Heptanoic acid, ester with 2,2-dimethyl-1,3-propanediol (CAS 68855-18-5) was performed, according to OECD Guideline 203 and GLP, in a semi-static system, using Oncorhynchus mykiss as test organism (Parr, 2012). During 96 h, the fish were exposed to the nominal test concentrations of 100% v/v, corresponding to a measured concentration of 0.086 mg/L. This was the highest attainable concentration, due to the poor solubility of the test substance. The test solutions were prepared using a solvent, and undissolved test material was removed by filtration. No mortality occurred during the study, and the LC50 (96 h) was reported to be > 0.086 mg/L.

Similarly, the second study 3,5,5-trimethylhexanoic acid mixed tetraesters with PE and valeric acid (CAS 131459-39-7) was performed, according to OECD Guideline 203 and GLP, in a semi-static system, using the same species as test organism (Wetton & Mullee, 1999). During 96 h, the fish were exposed to a loading test concentration of 100 mg/L, corresponding to a measured concentration of < 0.02 mg/L (below LOQ). The test solutions were prepared using a solvent, and undissolved test material was removed by filtration. No mortality occurred during the study, and the LL50 (96 h) was reported to be > 100 mg/L (nominal).

Based on the available results from these structurally related read-across substances (in accordance to Regulation (EC) No 1907/2006 Annex XI, 1.5) which are characterized by a similar ecotoxicological profile and comparable structure, it can be concluded that of 1,3-Propanediol, 2,2-dimethyl-, C5-9 carboxylates (CAS 85711-80-4) will not exhibit short-term effects on fish up to the limit of water solubility.
