Guideline:
OECD Guideline 201 (Alga, Growth Inhibition Test)
Deviations:
no
Qualifier:
according to guideline
Guideline:
EU Method C.3 (Algal Inhibition test)
Deviations:
no
GLP compliance:
yes (incl. QA statement)
Analytical monitoring:
yes
Details on sampling:

no
Test organisms (species):
Desmodesmus subspicatus (previous name: Scenedesmus subspicatus)
Details on test organisms:
TEST ORGANISM
- Common name:Desmodesmus subspicatus (formerly known as Scenedesmus subspicatus)
- Strain: CCAP 276/20
- Source (laboratory, culture collection): Laboratory culture (originally sourced from the Culture collection of Algae and Protozoa (CCAP), Scotland)
- Method of cultivation: In culture media in conical flasks with an initial cell density of approximately 10³ cells/mL

ACCLIMATION
- Acclimation period: Not stated
- Culturing media and conditions (same as test or not):The culture medium used for both the range-finding and definitive tests was the same as that used to maintain the stock culture
- Any deformed or abnormal cells observed: None
Test type:
static
Water media type:
freshwater
Limit test:
no
Total exposure duration:
72 h
Test temperature:
24 ± 1°C
pH:
7.6 - 7.9
Nominal and measured concentrations:
Analysis of the test preparations at 0 and 72 hours showed measured concentrations of manganese to be less than the limit of quantitation (LOQ) of the analytical method employed. This does not infer that no test material was in solution, just that that which was present was at a concentration below the LOQ.
Given that the toxicity cannot be attributed to a single component or a mixture of components but to the test material as a whole the results were based on nominal loading rates only.

Details on results:
- Exponential growth in the control (for algal test): yes

Reported statistics and error estimates:
One way analysis of variance incorporating Bartlett's test for homogeneity of variance (Sokal and Rohlf 1981) and Dunnett's multiple comparison procedure for comparing several treatments with a control (Dunnett 1955) was carried out on the growth rate and yield data after 72 hours for the control and all test loading rates to determine any statistically significant differences between the test and control groups.

Range finding test:

The cell densities and percentage inhibition of growth values from the exposure of Desmodesmus subspicatus to the test material during the range-finding tests showed no effect on growth at 0.010 mg/L loading rate WAF. However, growth was observed to be reduced at 0.10, 1.0, 10 and 100 mg/L loading rate WAF

From the data , it is clear that the growth rate (r) and yield (y) and of Desmodesmus subspicatus (CCAP 276/20) were affected by the presence of the test material over the 72 -hour exposure period.

Validity criteria fulfilled:
yes
Conclusions:
The effect of the test substance on the growth of Desmodesmus subspicatus has been investigated over a 72-hour period and gave the following results:
ErL50 = 90 mg/L (Loading rate WAF)
EbL50 = 39 mg/L (Loading rate WAF)
NOAEL = 10 mg/L (Loading rate WAF) (both biomass and growth rate)
Executive summary:

A study was performed to assess the effect of the test material on the growth of the green alga Desmodesmus subspicatus. The method followed that described in the OECD Guidelines for Testing of Chemicals (2006) No 201, "Freshwater Alga and Cyanobacteria, Growth Inhibition Test" referenced as Method C.3 of Commission Regulation (EC) No 440/2008.

Following both preliminary range-finding tests and initial experiments, Desmodesmus subspicatus was exposed to Water Accommodated Fractions (WAFs) of the test material over a range of concentrations of 1.0, 3.2, 10, 32 and 100 mg/L (three replicate flasks per concentration) for 72 hours, under constant illumination and shaking at a temperature of 24 ± 1°C.

Samples of the algal populations were removed daily and cell concentrations determined for each control and treatment group, using a Coulter® Multisizer Particle Counter.

Under the conditions of the study the 72-hour ErL50 was determined to be 90 mg/L loading rate WAF; the 72 -hour NOEL was determined to be 10 mg/L loading rate WAF.

Given that the toxicity cannot be attributed to a single component or a mixture of components but to the test material as a whole the results were based on nominal loading rates only.
