- The test substance was directly weighed into the test vessels (300 ml BOD bottles). The test substance was added to 234 mL of deionized water in a quantity appropriate for the desired concentration of 10, 100 and 1000 mg/L. Sixteen mL of synthetic sewage feed and 250 mL of microbial inoculum (activated sludge) were added to each test bottle.

- A sample of activated sludge was obtained from the Brazos River Authority in Sugar Land, TX and aerated when during transport and storage. The sludge was washed three times, with a settling time of 20 minutes between each wash.

- The Synthetic sewagerequired for the study was prepared using 16 gm peptone, 11 gm meat extract, 3 gm urea, 0.7 gm sodium chloride, 0.4 gm calcium chloride dthydrate, 0.2 gm magnesium sulfate heptahydrate, and 2.8 gm dipotassium hydrogen phosphate in 1 L of deionized water.
All test procedures took place in thoroughly cleansed 300 mL BOD bottles. The control, test, abiotic control and reference sample solutions were measured for dissolved oxygen content independently in series such that each bottle was measured immediately after the expiration of the three hour contact time. - Reference substance (positive control):
