Sampling and analysis
- Analytical monitoring:
- yes
- Details on sampling:
- Samples were taken after 0h, 24h, 48h and 72h and the concentration of the test item was determined.
Test solutions
- Vehicle:
- no
- Details on test solutions:
- The test concentrations were prepared by diluting a stock solution of 1 g/L. 200 mg test substance was weighed into a beaker, mixed with culture medium and was stirred for 5 min. with a magnetic stirrer to dissolve the test substance in the test medium. Then the test substance preparation was filled into a graduated flask and made up to a total volume of 200 mL with culture medium.
The amounts corresponding to the test concentrations were pipetted into a graduated flask, made up to a total volume of 1000 mL with culture medium, and then transferred to the test vessels (for dilution scheme, see table below).
Test organisms
- Test organisms (species):
- Desmodesmus subspicatus (previous name: Scenedesmus subspicatus)
- Details on test organisms:
- The unicellular algae Desmodesmus subspicatus was used as test organism. The organism (Strain 86.81) was obtained from “Sammlung von Algenkulturen”, Göttingen (SAG), (Collection of Algae Cultures, Institute for Plant physiology), University of Göttingen, Germany.
Three days before the start of the main experiment two flasks with 100 ml culture medium were prepared and 5 x 105 algae from stock culture were added, resulting in a final concentration of 5 x 103 cells per mL. After three days of incubation the cell concentration was determined by particle counter and was at least 16 x 5000 cells per mL. The mean of 5 measurements was calculated.
For the test an appropriate volume of the pre-culture with an amount of 7.5*10 000 cells per ml were used.

Applicant's summary and conclusion
- Validity criteria fulfilled:
- yes
- Conclusions:
- In an algal growth inhibition test using Desmodesmus subspicatus as test species, the test substance showed no effects on growth rate after 72h exposure up to concentrations of 25 mg/l (NOEC). The EC50 is considered to be > 100 mg/L (calculated as 255.8 mg/L) under conditions tested.
