The results as presented by Calamari and co-workers in 1980 indicate that tert-Butylamine is acutely harmful to fish if the test is performed in soft water (hardness: 20 mg CaCO3/L). The LC50 based on measured test item concentrations was determined to be 28 mg/L. The acute toxicity is reduced to a LC50 value of 270 mg/L if the test is performed in hard water with a hardness of 320 mg CaCO3/L. The low LC50 value for fish in soft water might be based on a shift in pH, since the low concentration of CO3- -ions in the test solution did not provide significant buffering capabilities. Unfortunately data on pH values in the test solutions are not presented in the publication, only a 'strong basic dissociation' is mentioned in the context of fish exposed to the test substance in soft water.

The hazard assessment is based on the LC50 value derived from the test using hard water since it is commonly accepted, that within the European Union, surface waters generally have a rather high hardness value. Therefore the LC50 determined in soft water is of low relevance.
