Sampling and analysis

Analytical monitoring:
yes
Details on sampling:
Samples were taken from freshly prepared control and test media samples at 0, 24, 48 and 72 hours, samples of the corresponding 24 hour aged media were taken after 24, 48, 72 and 96 hours.

Test solutions

Vehicle:
no
Details on test solutions:
A stock solution was prepared daily, 847 uL (or 424 uL) of the test substance was added to 10 L (or 5 L) of dilution water and mixed for two minutes with a magnetic stirrer. Following stirring any undissolved particles were removed by filtration through a 7 - 12 um pore size filter paper. The pH of the stock solution was 7.8. The test media were prepared by addition of appropriate volumes of the stock solution to test vessels and dilution to 5000 mL with water. A single replicate was used for each test treatment and contained seven fish.

Test organisms

Test organisms (species):
Danio rerio (previous name: Brachydanio rerio)
Details on test organisms:
The fish were obtained from Interaquaristic.de on 23 October 2009 and acclimated to the test conditions until used for the study. Fish were fed daily until 24 hours prior to the start of the study. The standard length of the fish was 3.41 cm with a standard deviation of 0.16 cm, n = 35

Study design

Test conditions

Hardness:
Dilution water - 250 mg/L as CaCO3
Test temperature:
Temperature range during the study was 20.6 - 21.7 °C. The temperature measured in the control vessel at the end of the end of the exposure period was 19.4°C, this was the only time the temperature fell below 20°C. This minor deviation is not condiered to have had nay effect on the outcome of the study.
pH:
pH range during the study was 7.5 - 7.8
Dissolved oxygen:
Dissolved oxygen (as % air saturation value) range during the study was 75 - 99%
Salinity:
Not applicable
Nominal and measured concentrations:
0 (control), 9.4, 20.7, 45.5 and 100 mg/L nominal concentrations. Geometric mean concentrations were determined for each 24 hour exposure period for each test concentration, the results are presented in the results section.
Details on test conditions:
A semi-static study was performed with renewal of the test substance every 24 hours.
Reference substance (positive control):
no

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Conclusions:
The 96 hr LC50 based on the geometric mean measured concentrations was 1.39 mg/L, the corresponding no-observed effect concentration (NOEC) was observed to be 0.501 mg/L.
Executive summary:

Zebra fish were exposed to nominal 2,3-dichlorobutadiene-1,3 concentrations of 0 (control), 9.4, 20.7, 45.5 and 100 mg/L for 96 hours under semi-static conditions according to EU Method C.1 Acute toxicity to fish (2008). Analysis of new and 24 hour aged media was performed for each treatment and geometric mean values were calculated for each 24 hour exposure period. The 96 hr LC50based on the geometric mean measured concentrations was 1.39 mg/L, the corresponding no-observed effect concentration (NOEC) was observed to be 0.501 mg/L.
