PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)

PREPARATION of test solution
-Preliminary test: prior to range-finding test, solubility test was conducted to identify the solubility of test in dilution water. As a result of solubility test, it's identified that the test substance is soluble with dilution water at the concentration of 500 mg/L.
-Range finding and limit test: the range finding and limit test are conducted at the nominal concentration of 10, 50, 80 and 100 mg/L. Each test solution of nominal concentration was prepared by dilution of stock solution in the morning of exposure date.
-Definitive test: the definitive test be conducted at the nominal concentration of 100 mg/L on the basis of range finding and limit test. Each test solution of nominal concentration was prepared by dilution of stock solution in the morning of exposure date.

TEST method: Homogeneity and stability of test substance were carried out by preliminary test according to the SOP of the test facility. The results of stability test, the concentration changes of test substance in nominal concentration (10 and 100 mg/L) after 96h were 1.1 and 12.6%, respectively. On the basis of the stability, the definitive test was carried out using static-system.

The range of body weight was 0.11 to 0.27g and that of total length was 2.5 to 2.9 cm in the representative test fishes.

Executive summary:

The study was performed to assess the acute toxicity effect of test item in fish (Oryzias latipes). according to the OECD Guideline for Testing of Chemicals No. 203 (1992).

 

1)      The test solution was carried out using static system throughout the duration of test.

2)      There were no abnormal appearances in all the test group at the end of the test.

3)      The body weight were 0.11~0.27 g and total length were 2.5~2.9 cm in the test fish.

4)      During the test period, temperature, pH and dissolved oxygen concentration of the test solutions were 22.1~23.1°C, 7.38~8.01 and 97.3~106.0%, respectively.

5)      Hardness of the test solution were 64~68 mg/L (as CaCO3).

6)      All the test solution were kept from the beginning to the end of test.

7)      The geometric mean of measured concentration for the test substance in the nominal concentration (100 mg/L) was 111.666 mg/L.
