Description of key information

Algal biomass and growth inhibition (species: Pseudokirchneriella subcapitata) is evaluated during 72 hours of exposure to Proviplast 0142, using number of cells as growth indicator. Test was performed according to OECD TG 201 Test concentrations were 0.62, 1.85, 5.56, 16.67 and 50 mg/l and no solvent was used. The results of the study showed that Proviplast 0142 has inhibitory effects on the biomass and growth rate of Pseudokirchneriella subcapitata. The ErC50 is 21.22 mg/l and EbC50 is 8.71 mg/l. The 72h-EC10 for growth rate and biomass are 7.05 and 2.32 mg/l respectively.
The estimated 96-hr LC50 value for algae for bis(2-butoxyethyl) adipate is 2.314 mg/l (ECOSAR v1.10). The estimated chronic value (ChV) is 1.059 mg/l, the corresponding NOEC is 0.749 mg/l. The acute and chronic toxicity is overestimated with a factor of 10 by the model.

Key value for chemical safety assessment

EC50 for freshwater algae:
21.22 mg/L
EC10 or NOEC for freshwater algae:
7.05 mg/L

Additional information

The effect of Proviplast 0142 on growth of the unicellular algal species Pseudokirchneriella subcapitata was measured during a 72 hours exposure period. Based on the results from a range finding study, the main study was conducted at 0.62, 1.85, 5.56, 16.67 and 50 mg/LProviplast 0142 in OECD TG 201 medium. No solvent was used. The test substance was not stable, with recovery below 80% at the end of the test. Results are expressed in geo mean values.

The test was performed in triplicate, at 22.6 to 23.4°C, pH 7.42-8.17 and light intensity of approximately 6774-6931 lux. The medium composition is reported.

At 0, 24, 48 and 72 hours the number of cells was measured in each test vessel (hemacytometer). Results were expressed as % growth inhibition compared to the control, for biomass and growth inhibition. The effect concentrations and the 95% confidence levels were calculated. Information to evaluate the test quality is available and the pH did not shift more than 1 unit. Test results are therefore valid.

Analytical results are documented for concentration and (in)stability of the test substance during the exposure.

The ErC50is 21.22 mg/l and EbC50is 8.71 mg/l. The 72h-EC10 for growth rate and biomass are 7.05 and 2.32 mg/l respectively. These values can be used for risk assessment.
