Sampling and analysis

Analytical monitoring:
yes
Details on sampling:
Samples for possible analysis were taken from all test concentrations and the control according to the schedule as follows:

Frequency: At 0 and 48 hours (start and end of test)
Volume: 3 mL from the approximate centre of the test vessels
Storage: Samples were stored in a freezer (≤-15°C) until analysis at the analytical laboratory of the Test Facility

At the end of the exposure period, the replicates were pooled at each concentration before sampling. Additionally, reserve samples of 3 mL were taken for possible analysis. If not used, these samples were stored in a freezer (≤-15°C).

Test solutions

Vehicle:
no
Details on test solutions:
PREPARATION AND APPLICATION OF TEST SOLUTION:
The batch of the test item was a clear yellow viscous liquid UVCB and not completely soluble in test medium at the loading rates initially prepared. No correction was made for the purity/composition of the test item. Preparation of test solutions was performed under dimmed light conditions with glassware wrapped in aluminium foil to minimize exposure to light.

Preparation of test solutions started with loading rates individually prepared at 1.0, 10 and 100 mg/L. A 31-minute period of ultrasonic waves followed by an overnight period of magnetic stirring was applied to ensure maximum dissolution of the test item in test medium. The obtained mixtures were centrifuged at 10000 rpm for 10 minutes. Subsequently, the supernatant was drained from the pelleted undissolved material using a pipette and the obtained Water Accommodated Fractions (WAFs) were used as test concentrations. All test solutions were clear and colorless at the end of the preparation procedure.

Details on results:
Measured concentrations:
Samples taken from the highest test concentration were analysed. The measured concentration was 46 mg/L at the start of the test. During the exposure period, the concentration remained stable, i.e. was at 112% relative to initial at the end of the test. Based on these results, effect parameters were expressed as the initially measured concentration and the loading rate. This measured concentration was determined to be the maximum solubility of the potentially soluble constituents of the UVCB substance. As such the EC50 values exceeded the limit of solubility.

Immobility:
No immobility was observed in the control and at the two lowest test concentrations throughout the exposure period. At the end of the test, 30% immobility was observed at the highest concentration tested.
Results with reference substance (positive control):
- Results with reference substance valid? Yes
- Relevant effect levels: 48 hour EC50 was 0.59 mg/L (with the historical range of 0.53 to 0.72 mg/L).
- Limit test: No
- Dose-response test: Yes (concentrations of 0.1, 0.18, 0.32, 0.56, 1.0 and 1.8 mg/L).
- ECx: EC50
Reported statistics and error estimates:
Not performed as EC50 greater than the highest concentration/loading rate tested.

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Conclusions:
In conclusion the 48 hour EL50 for Daphnia magna exposed to the test substance was greater than the highest loading rate tested (100 mg/L). Based on analytical measurements conducted of dissolved constituents the EC50 was greater than 46 mg/L (initial measured concentration). This was determined to be the the limit of solubility for the dissolved fraction.
