Description of key information

The partition coefficient (Log Pow) of Safranitrile was determined to be 3.4.

The partition coefficient of the test item, Safranitrile was determined according to OECD guideline 117, HPLC method according to GLP.

The method employed seven internal reference standards with known octanol/ water partition coefficient values within the range estimated for the test substance. The partition coefficient values for the reference substances ranged from log Pow 1.1 to log Pow 4.2. The log Pow of the test substance was determined to be 3.4, thus the reference substances were within the applicable range for determining log Pow for the test substance. A calibration mixture of the 7 reference substances was prepared and the retention times measured and averaged, and the decimal logarithms of the capacity factors (k) calculated using the deadtime as the retention time for thiurea. The capacity factor(k) was calculated to be 2.078. Log k was calculated to be slope (2.7609) × capacity factor + y axis intercept (2.5341). The linear regression parameters of the relationship log Pow= f(log k) were also calculated. The determination was performed at 35°C (temperature of the column), but the value obtained is considered to represent the value at an unspecified, probably ambient temperature since no mention is made in the guidelines of the temperature at which the reference substances log Pow values were determined. Partition coefficients of the test and reference substances are assumed to vary with temperature in the same way.
