Description of key information

The toxicity to microorganisms of the substances within the glycol esters category was assessed by read-across based on the grouping of substances (category approach). All available data were used for the assessment of the toxicity to microorganisms for all substances within the glycol esters category. The substances are readily biodegradable and in the toxicity controls of the biodegradation studies no inhibition of the degradation by the glycol esters substances was observed.

One experimental study investigating the toxicity of decanoic acid, mixed diesters with octanoic acid and propylene glycol (CAS 68583-51-7) to microorganisms is available which resulted in no inhibition of the respiration rate of Pseudomonas putida (EC50 > 0.99 g/L) (Diefenbach, 1995). This study was rated reliability 3 since it does not meet important criteria of today’s standard methods. Therefore, supporting data from the structurally related read-across substance butylene glycol dicaprylate / dicaprate (CAS 853947-59-8) were used according to Regulation (EC) No. 1907/2006 Annex XI, 1.5. to evaluate the toxicity to microorganisms. In this case of read-across, the best suited (highest degree of structural similarity, nearest physico-chemical properties) read-across substance was used for the assessment.

The study with the read-across substance butylene glycol dicaprylate / dicapratewas performed according to the EU Method C.11 (GLP) using domestic activated sludge as incoculum (Diefenbach, 1997). Five concentrations up to a maximum of 1099 mg/L were tested. After 3 h of incubation, no inhibition of the respiration rate was observed resulting in an EC10/50 > 1099 mg/L. The study performed with decanoic acid, mixed diesters with octanoic acid and propylene glycol was rated reliability 3, since a solubiliser was used for the preparation of test solutions. Using a solubiliser is not the appropriate method to use for complex mixtures. The use of a solvent can give preferential dissolution of one or more components and affect the toxicity (OECD, 2000). Nevertheless, no effects on the respiration rate of Pseudomonas putida were observed in this study (according to Hüls method under GLP conditions) after 4.75 h of incubation. Based on the results from a structurally related read-across substance (in accordance to Regulation (EC) No 1907/2006 Annex XI, 1.5) which is characterized by an equal ecotoxicological profile, it can be concluded that decanoic acid, mixed diesters with octanoic acid and propylene glycol will not exhibit effects on the respiration rate of microorganisms.
