Details on sampling:
- Concentrations: 0, 3.2, 5.8, 10, 18, 32 mg/L
- Sampling method: at 0 and 48 hours composite samples of each test concentration were drawn by mixing identical volumes of the test solutions taken from the approximate center of the test vessels.
- Sample storage conditions before analysis:kept at - 18 °C to - 22°C until analysis.
Vehicle:
no
Details on test solutions:
PREPARATION AND APPLICATION OF TEST SOLUTION
- Method: The stock solution was prepared by mixing 99.8 mg test substance with 400 mL water by polytron for 5 minutes and made up to 1000 mL with water. Calculated mounts of the stock solution to produce the desired test concentrations were given into the water and were homogeneously distributed. The daphnia were then transferred into the beakers. The test substance was homogeneously distributed in the test vessels at all test times and test concentrations.
Test organisms (species):
Daphnia magna
Details on test organisms:
TEST ORGANISM
- Common name: water Flea
- Strain/clone: STRAUS 1820
- Source: CIBA-GEIGY Ltd.
- Feeding during test: None during the test.
- Food type: A suspension of green algae (Scenedesmus subspicatus) supplemented by a suspension of TETRAMIN-extract.
- Amount: in such quantities that the food is consumed after 24h
- Frequency: 3x/week

ACCLIMATION
- Acclimation period: 24 hours before the begin of the exposure reproductive daphnia are separated from the young by sieving all individuals through a 800 µm sieve. This operation is repeated immediately before the exposure and the young (O-24h of age) are retained for the test.
- Breeding conditions (same as test or not): Cultures of daphnia are maintained in glass vessels containing approx. 2.5L of reconstituted water at 20 ± 1 °C (water is renewed partially thrice weekly). At each renewal the daphnia are fed.

TEST MEDIUM / WATER PARAMETERS
- Source/preparation of dilution water: Reconstituted water containing 65 mg NaHCO3, 294 mg CaCl2 * 2H2O, MgSO4 * 7H2O, 6 mg KCL 1000 mL bidistilled water.

OTHER TEST CONDITIONS

*Remark: The test substance is reported to be unstable when exposed to light. Therefore the exposure of the daphnia was done in darkness. As only 31-43% of the nominal concentration could be found at the end of exposure the EC and NOEC values are based on measured concentrations at 48 h.

The LC-50/EC-50 values were calculated according to the maxirnun likelihood method, probit model (Mc Cullagh, P., Nelder, J.A., 1983: Generalized linear models, Chapman & Hall, London)

Executive summary:

An acute aquatic toxicity test was performed according to OECD TG 202 (1984) and in compliance with GLP. 20 Daphnia magna (4 replicates, 5 organisms per test vessel) were exposed in a static system to nominal concentrations of 0, 3.2, 5.8, 10, 18, 32 mg/L. The test substance was reported to be unstable when exposed to light. Therefore the exposure of the daphnia was done in darkness. Measured 48h concentrations were <0.1, 1.0, 2.5, 3.7, 6.6, and 10.9 mg/L. Observations regarding mobility of the Daphnids were made at 24 and 48 hours. Based on final measured concentrations the 48-hour EC50 was calculated and graphically determined to be 2.8 mg/L. The 48 -hour NOEC and EC100 were calculated to be 1.0 mg/L and 6.6 mg/L, respectively.
