Reference
Endpoint:
short-term toxicity to fish
Type of information:
experimental study
Adequacy of study:
key study
Study period:
1990
Reliability:
2 (reliable with restrictions)
Rationale for reliability incl. deficiencies:
other: A OECD GLP 203 guideline study, but with standards from 1990. Analytical measurment of substance concentration not included, and there is no certificate of analyses.
Qualifier:
according to guideline
Guideline:
OECD Guideline 203 (Fish, Acute Toxicity Test)
GLP compliance:
yes (incl. QA statement)
Analytical monitoring:
no
Vehicle:
yes
Test organisms (species):
Poecilia reticulata
Details on test organisms:
The guppies were obtained from a aquarium retailer and were hold in stock for 10 days at the Research test lab. The size of the fish were approximatly 2 cm.
Test type:
semi-static
Water media type:
freshwater
Total exposure duration:
96 h
Hardness:
The diluting water was a synthetic water, Dutch Standard Water according to NPR 6507 (1980), having a ph of approximately 8.2 and a hardness of 13 dH, containing per litre of deionized water:
100 mg NaHCO3/L
20 mg KHCO3/L
200 mg CaCl2*H20/L
180 mg MgSO4*7H20

The deonized water contained not more than 0.01 mg copper per litre and had a conductivity of less than 5 µS/cm.
Test temperature:
24.5 - 25.5°C
pH:
8.2
Dissolved oxygen:
6.5-7,7 mgO2/L
Nominal and measured concentrations:
Only nominal values corrected for active ingredients are reported in this study.
Details on test conditions:
TEST SYSTEM
- Test vessel: 2Liters glass aquaria which contained 1 liter of test medium
- Renewal rate of test solution (frequency/flow rate): Semistatic test renewal ater 48 hours.
- No. of organisms per vessel: 10. In one of the test concentrations, one fish was unintentionally killed during removal of the test medium on day 2, and therefore the LC50 values for that concentration (18.9 mg/L) was calculated with 9 fish from they 0.
- No. of vessels per concentration (replicates): 1
- No. of vessels per control (replicates): 1
- No. of vessels per vehicle control (replicates) 1

TEST MEDIUM / WATER PARAMETERS
- Source/preparation of dilution water: Dutch Standard Water
- Total organic carbon: The TOC-content was not more than 2.0 mg/L.
- Conductivity:less than 5 µS/cm.
- Intervals of water quality measurement: 48 hours

TEST CONCENTRATIONS

- Range finding study: The selected range of the test concentrations were determined in two preliminary range finding tests with 10-100 mg/L and 4.2 - 9.0 - 18.9 - 41.4 mg/L
- Test concentrations: 18.9 -41.4 and 90 mg/L
Key result
Duration:
96 h
Dose descriptor:
LC50
Effect conc.:
47.7 mg/L
Nominal / measured:
nominal
Conc. based on:
act. ingr.
Basis for effect:
mortality (fish)
Remarks on result:
other: 95 % CL: 17.3 -131.6 mg/L
Reported statistics and error estimates:
In one of the test concentrations, one fish was unintentionally killed during removal of the test medium on day 2, and therefore the LC50 values for that concentration (18.9 mg/L) was calculated with 9 fish from they 0.
Validity criteria fulfilled:
yes
Remarks:
Mortality in control <10%, the oxygen concentration 60% of air saturation.
Executive summary:

This OECD 203 study performed under GLP conditions resulted in a LC50 of cyclohexanone peroxide to Guppy of 47.7 mg/L.The study meets the relevant validation criteria.
